摘要
目的:探讨雄黄对卵巢癌细胞株SKOV3和COC1增殖能力的影响及可能机理。方法:采用不同浓度雄黄溶液作用于两珠细胞,用MTT法检测生长抑制率;流式细胞仪检测细胞凋亡率及分析细胞周期;原位末端标记法(TUNEL)观察细胞凋亡形态学。结果:不同浓度的雄黄溶液对卵巢癌细胞株SKOV3及COC1均有不同程度的生长抑制作用,具有浓度和时间依赖性,且COC1比SKOV3对雄黄更敏感。结论:雄黄对卵巢癌细胞有生长抑制作用,诱导细胞发生G1期阻滞是雄黄抑制卵巢癌细胞生长作用的可能机制之一。
Objective:To study the effecs of realgar on proliferation of SKOV3 and COC1 cell lines.Methods: Under different concentration of Realgar solution,the inhibition effect of realgar on the cell proliferation of ovarian carcinoma cell lines was tested using methyl thiazolyl tetrazolium(MTT) assay.The apoptosis percentage and phase distribution of cell cycles were detected by Flow cytometry(FCM).After exposure to different concentration realgar solution,apoptosis morphologic features of SKOV3 and COC1 were observed by TdT-mediated –dUTP nick end labeling(TUNEL).Results:Different concentrations of realgar solution of SKOV3 and COC1 have varying degrees of growth inhibition in a concentration and time dependent.The growth inhibition of SKOV3 cells with realgar was weaker than that in COC1 cells.Conclusion:The growth of ovarian cancer cells are inhibited with treatment of different concentration realgar solution.Realgar can block cell cycle arrest at the G1 phase,which may be one of the mechanisms of apoptosis induced by Realgar.
