摘要
目的:探讨安宫牛黄丸对大鼠脑缺血后脑组织内热休克蛋白70(HSP70)表达的影响及意义。方法:线栓法制备大鼠局灶性脑缺血模型,将55只SD大鼠随机分为假手术组(5只)、缺血对照组(25只)、安宫牛黄丸治疗组(25只),缺血组和治疗组再各分为12h、24h、3d、7d、14d共5个亚组,每个亚组5只,治疗组大鼠给予安宫牛黄丸0.5g/2mL灌服,1天1次,直到各亚组时间点;对照组及假手术组同时予等量生理盐水灌服。观察各组大鼠脑梗死体积、组织形态学变化,并免疫组化方法检测HSP70蛋白含量。结果:安宫牛黄丸治疗组较缺血组梗死灶体积明显减少(P<0.05),缺血组HSP70阳性细胞逐渐增多,于3天达最高峰,然后逐渐降低。在缺血再灌注3天后开始,安宫牛黄丸治疗组HSP70阳性细胞较缺血对照组明显增多(P<0.05)。显微镜下见HSP70细胞主要表达于缺血半暗带区。结论:安宫牛黄丸治疗对大鼠局灶性脑缺血再灌注损伤有保护作用,其作用机制可能与增加HSP70表达有关。
Objective: To investigate the effectiveness of Angong Niuhuang Pill(ANGHP) on heat shock protein70 (HSP70) expression in rats with cerebral ischemia injury. Methods: The models with focal cerebral ischemia in rats were established by the suture occluded method. Fifiy-five SD rats were randomly divided into 3 groups: sham group(5), ischemia group(25),and AGNHP group(25). The ischemia group and AGNHP group were further di- vided into 5 subgroups:12h,24h,3d,7d, and 14d(5 in each subgroup). In the AGNHP group,rats were given gavage of AGNHP 0.Sg/2ml once a day until the ending time of each subgroup,whereas the sham group and the isch- emia group were given NS. The infarct volume of the rat brains in each group was determined,the neurons were observed under microscopy,and the expression of HSP70 was detected by immunohistochemistry. Results: The in- farct volume of the AGNHP group was reduced than that of the ischemia group with a significant difference(P〈 0.05). The HSP70 expression in the ischemia group was gradually increased to a peak in 3 days, and then turn to decrease. Starting from the 3rd day, the HSP70 expressions in the AGNHP groups was increased significantly as compared with that in the ischemia group(P〈0.05). The HSP70 cells mainly expressed in the ischemic penum- bra district. Conclusion: AGNHP could protect brain cell in rats with cerebral ischemia by increasing the expression of HSP70.
