PGC-1α与中枢性肥胖小鼠的机体能量代谢和肥胖发生的相关性 被引量:2

Relationship between peroxisome proliferator-activated receptor gamma coactivator 1alpha(PGC-1α)with body energy metabolism and obesity in central obese mice

摘要 目的通过观察过氧化物酶体增殖物活化受体γ共激活因子-1α(PGC-1α)在中枢性肥胖小鼠中的表达水平研究PGC-1α与能量代谢和肥胖的相关性。方法皮下注射谷氨酸钠建立小鼠中枢性肥胖的病理模型,比较各组小鼠的BMI、摄食量及测定体温的变化;检测PGC-1α在棕与白色脂肪、肝脏、肺脏、心脏和脑中的表达水平。结果 PGC-1α在上述组织中都有不同程度的表达;而模型组PGC-1α的表达下调。PGC-1α在棕色脂肪的表达始终高于白色脂肪;寒冷刺激可诱导脂肪组织中PGC-1α表达增加,且在棕色脂肪中尤为显著。结论 PGC-1α可能与机体能量代谢和肥胖的发生密切相关。 Objective To investigate the level of peroxisome proliferator-activated receptor gamma coactivator 1alpha(PGC-1α)expression in the central obese mice in order to get the relationship between PGC-1α with energy metabolism and obesity.Methods To create the obese models,the glutamic sodium was injected by subcutaneous.The body mass index,body temperature,food intake were examined.Besides,the level of PGC-1α expression in brown adipose,white adipose,liver,lung,heart and brain were detected through RT-PCR.Results The expression of PGC-1α in brown adipose,white fat,liver,heart,lung and brain were different.Compared with the normal group,the expression of PGC-1α was basically reduced in the model group.In addition,the PGC-1α expression in white and brown adipose were up regulated in cold stimulation and more significantly increased in brown adipose.Conclusion It′s likely that PGC-1α was associated closely with body energy metabolism and obesity.
