摘要
[目的]从分子水平阐述大鼠FSHβmRNA表达的GnRH受体后信号转导机制。[方法]采用细胞体外培养技术结合实时荧光定量PCR技术,检测大鼠GTH细胞中cAMP改变对FSHβmRNA表达的影响。[结果]cAMP含量对大鼠GTH细胞表达的FSHβmRNA的Ct值的影响不显著。FSK和SQ22536能有效改变GTH细胞的cAMP含量,却不能改变PKC活性。FSHβ的表达是受GnRH调控的。[结论]cAMP不是FSHβmRNA表达的GnRH受体后的信号转导途径。
[Objective] The research aimed to discuss the GnRH post-receptor signal transduction mechanism of the expression of FSH β mRNA in rats at molecular level.[Method] Using cell culture technique in vitro and real-time fluorescence quantitative PCR technology,the effects of cAMP changes in GTH cells on the expression of FSHβ mRNA in rats were studied.[Result] cAMP content had no significant effect on Ct value of FSHβ mRNA expressed by GTH cells in rats.FSK and SQ22536 could effectively change cAMP content of GTH cells,but it could not change PKC activity.The expression of FSHβ was regulated by GnRH.[Conclusion] cAMP is not GnRH post-receptor signal transduction pathway for the expression of FSHβ mRNA.
