摘要
目的建立同时测定心可舒颗粒中主药有效成分葛根素和芦丁含量的方法。方法采用Symmetry C18色谱柱(250 mm×4.6 mm,5μm),流动相为甲醇-水-乙腈-冰醋酸(28∶60∶10∶2),流速为0.8 mL/min,检测波长为252 nm,柱温为30℃。结果葛根素质量浓度在2.54~25.40μg/mL的范围内与峰面积呈良好线性关系(r=0.999 8),平均回收率为98.64%,RSD=2.25%(n=6);芦丁质量浓度在1.97~19.70μg/mL范围内与峰面积线性关系良好,r=0.999 6,平均回收率为100.57%,RSD=2.53%(n=6)。结论该法准确、简便,重现性好,可用于心可舒颗粒制剂的质量控制。
Objective To establish a HPLC method for the determination of puerarin and rutinum in Xinkeshu Granules.Methods The analytical column was the Symmetry C18 column(250 mm×4.6 mm,5 μm) with methanol-water-acetonitrile-glacial acetic acid(28:60:10:2) as the mobile phase.The flow rate was 0.8 mL/min,the detection wavelength was 252 nm and the column temperature was 30 ℃.Results The linear range for puerarin and rutinum were 2.54-25.4 μg/mL(r=0.999 8) and 1.97-19.7 μg/mL(r=0.999 6),respectively.The average recovery rates were 98.6%,RSD=2.23%(n=6) for puerarin and 100.5%,RSD=2.08%(n=6) for rutinum.Conclusion This method is simple,accurate,better reproducible,and suitable to be used in the quality control of Xinkeshu Granules.
