参麦及顺铂对人胃癌SGC-7901细胞生长的协同抑制作用 被引量:2

The synergistic effect of shenmai and cisplatin on the growth of the human gastric cancer cell SGC-7901

摘要 目的探讨参麦及顺铂对人胃癌SGC-7901细胞生长的协同作用及其可能机制。方法体外培养人胃癌SGC-7901细胞,MTT法测定细胞增殖情况;Hoechst33258染色观察细胞凋亡形态学变化;RT-PCR检测VEGF、Bcl-2和Caspase-3mRNA的表达。结果 MTT法显示参麦及顺铂能抑制胃癌SGC-7901细胞增殖,其作用具有时间及浓度依赖性,单用顺铂组,有效浓度为1.25μg/ml,起效时间为72h,其抑制率为(8.70±0.57)%,联合用药组,有效浓度为顺铂0.625μg/ml,起效时间为48h,其抑制率为(8.87±0.10)%。行Hoechst33258染色可见细胞核出现固缩、边集,形成凋亡小体等凋亡现象;RT-PCR检测显示:与对照组相比,药物作用后Bcl-2、VEGF的mRNA表达降低,Caspase-3mRNA表达升高。结论参麦与顺铂联合对胃癌细胞的抑制具有协同作用,与单用顺铂相比,参麦与顺铂联合可有效减少顺铂的用药剂量,促进凋亡,其作用与协同促进Caspase-3的高表达及Bcl-2、VEGF的低表达有关。 Objective To evaluate the synergistic effect of shenmai plus cisplatin (DDP)on the human gastric carcinoma cell SGC-7901 and underly the mechanisms involved in. Methods SGC-7901 gastric cancer cells were cultured with DDP alone or in combination with shenmai in vitro. Drug inhibitory effect on proliferation of cells were measured by MTF assay. Morphological changes of cells were observed by Hoechest33258 staining. The expression of Bcl-2, Caspase-3 and VEGF were detected by reverse-transcription PCR (RT-PCR). Results The proliferation of SGC-7901 was inhibited significantly by DDP in combination with shenmai in concentration- and time- dependent manner by MTY assay. In DDP alone group, the effective concentration was 1.25 μg,/ml, and the onset time was 72 h, while its inhibition ratio was ( 8. 70 ± 0. 57 ) %, however, in shenmai plus DDP group, the effective concentration was 0. 625 μg/ml, and the onset time was 48 h, inhibitory ratio in the group was ( 8.85 ± 0. 1,) %. Apoptotic features such as nuclear pyknosis, chromatin margination, and apoptotic bodies were observed by Hoechest33258 staining. Compared with the control group, the mRNA expression of Bcl-2, VEGF were decreased while that of Caspase-3 was increased in the group of DDP alone or in combination with shenmai. Conclusions Shenmai and DDP have synergistic effect on the inhibition of gastric carcinoma cells. The therapeutic dose of DDP in combined treatment group was less than that in DDP group. Besides, the apoptosis was promoted by combined treatment. The synergetic effect associated with the up-regulation of Caspase-3 and down-regulation of Bcl- 2 and VEGF.
