血管内皮祖细胞介导的急性肺损伤的保护机制的研究 被引量:1

Research of the protection of acute lung injury mediated by endothelial progenitor cells

摘要 目的探讨体外培养的血管内皮祖细胞(EPCs)对急性肺损伤的保护作用。方法体外培养被荧光标记的EPCs,取80只BALB/C小鼠,平均分成4组,对照组(control组),急性肺损伤组(ALI组),急性肺损伤后注射EPCs组(ALI+EPCs组),无急性肺损伤注射EPCs组(EPCs组),四组分别在实验3d后同时处死后测肺水含量(EVLW)、肺通透指数(PPI),分别对其相关性进行观察和分析,并观察其肺组织冰冻切片的荧光表达。结果 ALI+EPCs组肺组织冰冻切片荧光表达EPCs量较EPCs组多,ALI组的术后肺组织EV-LW、PPI与control组和EPCs组相比有显著差异(P<0.05),ALI+EPCs组的术后EVLW、PPI与control组、ALI组和EPCs组均有显著差异(P<0.05),而control组和EPCs组的术后EVLW、PPI之间无显著差异(P>0.05)。结论体外培养的EPCs对损伤的肺血管内皮细胞有显著地趋化修复作用,因而对急性肺损伤有显著保护作用。 Objective To investigate the protection of endothelial progenitor cells which was cultured in vitro on acute injured lung. Methods Culturing endothelial progenitor cells carried fluorescent mark in vitro, 80 BALB/C mice were devided into four groups, control group, acute lung injury group (group ALI) , injecting EPC after acute lung injury group (group ALl + EPCs ), injecting EPC without acute lung injury group (group EPCs ), all mice were executed 3 days after experiments. The content of edema with the lung involved and index of pulmonary permeability were measured, lhe relativity between them were analyzed and the expression of the fluorescent of lung was observed. Results Fluorescent was more obvious on group ALl + EPCs than group EPCs. The content of edema with the lung involved and index of pulmonary permeability in group ALI were more significant different than control group and group EPCs. The content of edema with the lung involved and index of pulmonary permeability in group ALI+ EPCs were significant different than control group and group ALI and group EPCs, while in control group and group EPCs were not significant. Condusions Repair and protection of endothelial cells of acute injured lung of EPCs cultured in vitro is obvious, so the protection of acute lung injury of EPCs is obvious.
