摘要
报道了利用Al3+作为柱前衍生试剂,高效液相色谱分离荧光检测法检测测定四环素类药物的新方法。探讨了四环素类药物-Al3+络合物的衍生条件及其色谱分离条件对荧光强度的影响,3种药物的检出限达70-150pg。
A method is described for the determination of tetracyclines as complexes with Al3+ by reversed-phase high-performance liquid chIDInatogIahy and fluonence detection.The complexes of tetracycline-, chlortetracycline-and minocycline-Al3+ were derivatized in a solution containing buffer of pH=2. 9 and separated on a Kromasil ODS column(250 x 4.6 mm, I. D. 5μm) using acetonitrile-water (90:10, V/V) containing 0.025 mol/L Li2SO4 (pH=2.9) as mobile phase at a flow rate of 0.8 mL/min, and detect at λex = 383 nm andλex = 480 nm.The detection limits were 0.1 ng for tetracycline,0.15 ng for chlortetracycline and 70 pg for minocycline.The method has been applied to the determinations of eye unguent of chlortetracycline and cortison eye unguent of tetracycline with satisfactory results.
