HPLC法测定当归补血总苷中阿魏酸的含量

Determination of Ferullic acid in total glucosides of Danggui-buxue-tang by HPLC

目的建立RP-HPLC法测定当归补血总苷中阿魏酸含量的方法。方法 ODS C18柱为固定相(150×4.6 mm,5μm),乙腈-1%醋酸(17:83)为流动相,流速1.0 ml/min,检测波长:320 nm,柱温为30℃。结果阿魏酸的线性范围为3.3～26.4μg/ml(r=0.999 8),平均回收率为99.6%(RSD=1.6%)。结论本方法简便、快速,重现性好,结果准确可靠,可作为当归补血总苷的质控方法之一。

Objective To establish the method for determining Ferullic acid in total glucosides of Danggui - buxue - tang （TGDB） by HPLC. Methods RP - HPLC was adopted with Diamonsil C18 chromatographic column （ 150 × 4.6 mm,5 μm）, Methyl Cyanides - 1% acetic acid was applied as the mobile phase（ gradient elution）, flow rate was 1.0 ml/min, diode array detector was set at 320 nm and column temperature at 30℃. Results The calibration curve of the absorbency was linear with the concentration of Ferullic acid in the range of 3.3 × 26.4 μg/ml （r = 0. 999 8）. The average recovery of Femllic acid was 99.71% , and RSD was 1.61%. Conclusion The method is accu- rate, simple, fast and feasible and can be applied as one of quality control methods of total glucosides of Danggui - buxue - tang.
