摘要
目的优化幽门螺杆菌体外培养条件。方法通过幽门螺杆菌培养基处方及培养环境的对比,探索体外培养的实验综合条件。结果与结论用哥伦比亚培养基作为培养基础制成的选择性培养基培养效果较好,用四通管把培养罐、手动负压泵、压力表和混合气体瓶连接起来,采用抽气-换气法可以有效保证Hp所需的气体环境,并且经济实惠。幽门螺杆菌在添加了混合抗生素和优质胎牛血清的哥伦比亚培养基培养效果优胜于其它选择性培养基,实验的用品及操作过程中严格按照无菌操作规范,能有效地防止杂菌和霉菌的滋生,为提高幽门螺杆菌体外培养率提供条件。
Objective To optimize the conditions for in vitro cultivation of Helicobacter pylori (Hp). Methods Through the contrast of Hp medium formulation and culture environment, the comprehensive conditions for in vitro cultivation were explored. Results and Conclusions The selective medium based on the Columbia culture medium had the better culture effect. The connection of cultivating pot, manual negative pressure pump, pressure gauge and mixed gas bottle by four-way pipes can effectively guarantee the gas environment of Hp culture with economic benefits. The culture effect of Columbia culture medium combined with antibiotics and high-quality fetal bovine serum for Hp is better than other selective medium. The experimental supplies and operation process with aseptic practices can effectively prevent the breeding of bacteria and fungi, and provide the conditions for in vitro cultivation ofHelicobacter pylori.
