摘要
以小麦粒固体培养基栽培高雄山虫草。筛选了适于生产的优良菌株,该菌株栽培后生长良好,搔菌后出草较多、整齐,但生物量和生物转化率与未搔菌相比没有明显差异;搔菌组第二潮子实体生物量和生物转化率均优于未搔菌组,搔菌组平均每瓶分别为(0.33±0.01)g和(2.2±0.01)%,未搔菌组分别为(0.12±0.01)g和(0.7±0.01)%。经过二潮子实体培养总生物量和总生物转化率高于一次培养。
An anamorphic strain of Cordyceps takaomontana,GZUIFR-PT123,was selected for artificial cultivation based on the production of higher numbers of uniform(less branching) fruit bodies on potato dextrose agar.GZUIFR-PT123 mycelium was grown at 25 ℃ in culture bottles(13 cm high,6 cm diameter) containing a substrate mix consisting of 15 g wheat seeds,4 g silkworm chrysalis meal,0.5 g maltose,10 mL mineral salts solution(0.1% CaCl2,0.5% ZnSO4 and 0.5% MgSO4),5 mg vitamin B1and 25 mL water.After the growth medium was fully colonized(8~10 d),the upper surface(0.3~0.5 cm) was removed("scratching") and discarded.At the first flush,no significant differences in fruit body production were recorded between bottles subjected to "scratching" treatment and controls in which the upper surface was retained.However,after scratching,GZUIFR-PT123 fruit bodies were more uniform,and biological efficiency values at the second flush was significantly higher in treated bottles(2.2%) compared with untreated controls(0.7%).
