反相高效液相色谱法测定瓦尼木层孔菌中柚皮素的含量 被引量:1

摘要 采用反相高效液相色谱法(RP-HPLC)测定瓦尼木层孔菌(Phellinus vaninii)中柚皮素含量。瓦尼木层孔菌醇提物水解后采用RP-HPLC对柚皮素的含量进行测定,色谱条件:Hypersil ODS(4.6mm×100mm,3μm)色谱柱,流动相为1%冰醋酸溶液-甲醇(62∶38),柱温为40℃,流速为0.6mL/min,检测波长为288nm。标准曲线为A=4.18×106 X+40109(r=0.9998,n=5),柚皮素进样量在0.0202~2.020μg间线性关系良好,相对标准偏差(RSD)为1.56%(n=5),回收率为97.8%,重现性实验RSD为1.76%。该法适于瓦尼木层孔菌醇提物中柚皮素的含量测定。 A method for determining naringenin levels in Phellinus vaninii fruit bodies by reversed-phase highperformance liquid chromatography(RP-HPLC)is described.Ethanolic(70v/v)extracts of P.vaninii fruit bodies were treated with 2% H2SO4at 100℃for 30mins.After adjusting the pH to 7.0with 40% NaOH,the solution was extracted with ethyl acetate and the extracts were freeze-dried.Samples(400 mg)of the freeze-dried material were dissolved in 25mL methanol,aliquots(20μL)of the solution were applied to a Hypersil ODS chromatography column(4.6mm×100mm,3μm),and the column was eluted with a mobile phase consisting of a mixture of 1%(v/v)glacial acetic acid and methanol(62∶38,v/v).The column temperature and flow rate were 40 ℃ and 0.6 mL/min respectively,and the detection wavelength was 288nm.Naringenin values were linear over the range 0.0202~2.020μg(RSD = 1.47%-1.56%),and the average recovery rate was 97.8%(RSD=2.07%,n=5).
