摘要
目的探讨左归丸对免疫性卵巢早衰小鼠(POF)卵巢Bcl-2、Bax蛋白表达的影响。方法以小鼠透明带3为抗原,皮下多点注射免疫BALB/c雌性小鼠建立免疫性卵巢早衰模型。设左归丸低、中、高剂量进行治疗,以泼尼松、己烯雌酚为阳性对照,免疫组化法检测卵泡、卵巢间质Bcl-2、Bax蛋白的表达。结果与空白对照组比较,模型组大卵泡及小卵泡Bcl-2蛋白的表达均减少,Bax蛋白的表达均增加,差异有统计学意义(P<0.05)。与模型组相比,左归丸高剂量组大卵泡及小卵泡Bcl-2蛋白的表达明显增加,差异有统计学意义(P<0.001);左归丸高、中、低剂量组卵巢间质Bcl-2蛋白的表达均明显增加,差异有统计学意义(P<0.05~0.001),大卵泡及小卵泡Bax蛋白的表达均减少,差异有统计学意义(P<0.01~0.001);左归丸中剂量组卵巢间质Bax蛋白的表达减少,差异有统计学意义(P<0.05)。左归丸各剂量组卵泡Bcl-2蛋白的表达呈现一定的量效关系。结论卵泡凋亡过快可能导致卵巢早衰,左归丸可能通过上调卵泡及卵巢间质Bcl-2蛋白的表达,下调Bax蛋白的表达,抑制卵泡的过快凋亡,从而改善卵巢功能。
Objective To probe the effect of Zuogui Pill(ZGP), on expression of Bcl-2 and Bax protein in ovaries of mice with immune premature ovarian failure (POF). Methods BALB/c female mice model of POF was established by multiple sites subcutaneous injection of zona pellucida 3 of mice, and then were treated with low, middle and high dosage ZGP, or positive control medicine of prednisone and diaethylstilbestrol. Expression levels of Bcl-2 and Bax protein in mice folliculi and ovarian stroma were detected by immunohistochemical method. Results Compared to the black control group, the expression of Bcl-2 protein of great folliculi and small ones of model group decreased obvi- ously and the expression of Bax protein of great folliculi and small ones of model group increased obviously (P 〈 0.05 ). Compared to the model group, the expression of Bcl-2 protein of great folliculi and small ones of high-dosage ZGP group increased obviously(P 〈 0.001 ) ; the expression of Bcl-2 protein of ovarial stroma of low, middle and high dosage ZGP group increased obviously (P 〈 0.05 -0.001 ), the expression of Bax protein of great folliculi and small ones of different dosage decreased obviously(P 〈 0.01 - 0.001 ) ; the expression of Bax protein of ovarial stroma in the mice of middle dosage ZGP group decreased obviously (P 〈 0.05 ). All the data showed a dose-response relationship to some degree in expression of Bcl-2 protein within low, middle and high dosage ZGP group. Conclusion It is indicated that rapid apoptosis of follicle may result in premature ovarian failure, and ZGP can improve the function of ovaries by up regulating the expression of Bcl-2 protein and down regulating the expression of Bax protein in folliculi.
