HPLC测定消炎止痒醑中盐酸异丙嗪的含量

Determination of promethazine hydrochloride in Xiaoyan Zhiyang spirit by HPLC

目的:建立HPLC测定消炎止痒醑中盐酸异丙嗪的含量。方法:色谱柱为Xierra-RP18柱(4.6mm×150mm,5μm);流动相:甲醇-0.01mol/L磷酸二氢钠缓冲液(取1.56g磷酸二氢钠,加纯水1000ml溶解,用磷酸调节PH值至3.0)(50:50);流速:1.0mL/min;检测波长:251nm;进样量:5μL。柱温:30℃。结果:盐酸异丙嗪进样量在0.2002～1.2012μg范围内与峰面积呈良好的线性关系,r=0.9997,平均回收率为100.95%,RSD为0.54%(n=9)。结论:本测定方法简便、快速、准确、重复性好,可作为消炎止痒醑的含量测定方法。

Objective：To Establish a menthod for determination of promethazine hydrochloride in Xiaoyan Zhiyang spirit by HPLC.Methods：The Xierra-RP 18（4.6mm×150mm,5μm）column was used;The mobile phases consisted of methanol and 0.01mol/L sodium dihydrogen phosphate（1.56g sodium dihydrogen phosphate,add 1000ml of pure water solution,adjust PH to 3.0 by phosphoric acid） （50：50）;the flow rate was 1.0mL/min and the detection wavelength was at 251nm;the injection volume was 5μL.The column temperature was 30℃.Results：The peak area of promethazine hydrochloride showed a good linearity in the range of 0.2002～1.2012μg,r=0.9997.The average recovery was 100.95%,RSD=0.54%（n=9）.Conclusions：The method was simple,fast,accurate,reproducible and can be used as quality control for the Xiaoyan Zhiyang spirit.
