摘要
[目的]对布拉酵母菌(Saccharomyces boulardii)的高密度培养条件进行研究。[方法]以布拉酵母为试验菌株,通过分批发酵和流加培养筛,分别测定温度、pH、溶氧(DO)、起始葡萄糖浓度、残糖浓度及氨基氮浓度对布拉酵母生长代谢的影响。[结果]试验确定布拉酵母高密度培养的最佳工艺为:培养温度30~32℃,pH 5.0,溶氧30%,初始葡萄糖浓度30 g/L,培养过程中通过流加补料维持发酵液中葡萄糖浓度为0.5 g/L、氨基氮浓度为40 mg/L。在此条件下培养24 h后细胞干重最高可达56.6 g/L,细胞得率为42.4%。[结论]该研究对布拉酵母活菌制剂的生产制备具有重大参考价值。
[Objective] The aim was to study on the high-cell density culture conditions of Saccharomyces boulardii.[Method] With Saccharomyces boulardii as the test organism,over batch fermentation and fed batch culture,the effects of temperature,pH,dissolved oxygen(DO),initial glucose concentration,leftover glucose concentration and amino nitrogen concentration on the growth and metabolism of Saccharomyces boulardii were determined.[Result] The optimum process of high-cell density culture conditions of Saccharomyces boulardii was determined that the temperature was 30-32 ℃,pH 5.0,30% DO,initial glucose concentration was 30 g/L,and maintaining the glucose concentration of 0.5 g/L and the amino nitrogen concentration of 40 mg/L by fed-batch during culture.When cultured in the optimum conditions after 24 h,the dry cell weight was reached 56.6 g/L and cell rate was reached 42.4%.[Conclusion] It had significant reference value for the production and preparation of the Saccharomyces boulardii live bacteria preparation.
