摘要
目的探讨抗辐灵对微波辐射致大鼠精子损伤的防治作用。方法 100只Wistar雄性大鼠,随机分为:正常对照组、辐射对照组、1 g/(kg.d)抗辐灵组、2 g/(kg.d)抗辐灵组和4 g/(kg.d)抗辐灵组,30 mW/cm2微波全身辐射10 min,于辐射前7 d灌胃给予1、2和4 g/(kg.d)抗辐灵水溶液,每天1次,连续14 d。分别于停药后6 h(辐射后7 d)、7 d(辐射后14 d)、14 d(辐射后21 d)处死大鼠,SCA精子动态分析系统检测大鼠精子活力参数,HE染色观测精子畸形率。结果 30mW/cm2微波辐射后7 d,大鼠精子畸形率升高(P<0.05);停药后6 h各剂量药物组精子畸形率较辐射组明显降低(P<0.05或P<0.01)。30 mW/cm2微波辐射后14 d大鼠精子活动率(AR)和A+B级精子比例明显减少(P<0.05),D级精子比例明显增加(P<0.05),精子畸形率增加(P<0.05)。停药后7 d;2和4 g/(kg.d)抗辐灵组精子活动率(AR)和A+B级精子比例较辐射组明显增加(P<0.05或P<0.01),D级精子比例降低(P<0.05),精子畸形率降低(P<0.05)。结论 30 mW/cm2微波辐射可引起大鼠精子损伤,表现为精子活动率减少和精子畸形率增加,抗辐灵具有明显防治微波辐射损伤的作用。
Objective To study the effect of a traditional Chinese medicine " Kang Fu Ling" (KFL) on sperm injury in rats induced by microwave radiation. Methods One hundred male Wistar rats were divid- ed into 5 groups randomly: normal control (20 rats), microwave radiation control (20 rats) , 1 g/( kg· d) KFL (20 rats), 2 g/(kg· d) KFL (20 rats) and 4 g/(kg ·d) KFL (20 rats) groups. The rats were ex- posed to microwave radiation at an average power density of 30 mW/cm2 for 10 minutes. Intragastric KFL administration was given at the doses of 1, 2 and 4 g/kg/d in 7 days before microwave exposure and con-tinued for 14 days. Rats were sacrificed at 6 hours, 7 days and 14 days after the final intragastric Kang Fu Ling administration. Meanwhile, sperm motilities were detected with Sperm Class Analyzer (SCA) , and the ratio of sperm abnormality was observed with HE staining. Results Compared with the sham group, the ratio of sperm abnormalities of rats in the 30 mW/cm2 irradiation group was significantly increased on the 7th day after microwave irradiation (P 〈 0.05). Compared with the radiation group, the ratio of sperm abnormalities of rats in the 1, 2 and 4 g/kg/d KFL groups was decreased at 6 hours after the final intragastric drug administration (P 〈 O. 05 or P 〈 O. O1 ). The ratio of sperm activity and the percentage of grade A + B sperm of the rats were decreased in the 30 mW/cm2 group on the 14th day after microwave exposure (P 〈 0.05 ), while the percentage of grade D sperm increased (P 〈 0.05 ) and the sperm abnor- mality increased (P 〈 0.05). Compared with the radiation group, the ratio of sperm activity and the per- centage of grade A + B sperms in the 2 and g g/kg/d KFL groups were increased at the 7th day after the last intragastric administration (P 〈0.05 or P 〈 0.01 ), while the percentage of grade D sperm and the ratio of sperm abnormality was significantly decreased (P 〈 0.05). Conclusions Microwave radiation at the average power density of 30 mW/cm2 might damage rats' sperms, such as the reduction of sperm motility and increase of sperm abnormality. KFL exert preventive and therapeutic effect on injuries induced by microwave radiation.
