β-榄香烯对血管内皮细胞成血管能力、细胞凋亡及MMP-2和MMP-9活性的影响 被引量:14

摘要 目的观察β-榄香烯对血管内皮细胞成血管能力、细胞凋亡及基质金属蛋白酶2(MMP-2)和MMP-9活性的影响,探讨β-榄香烯对肿瘤血管生成的抑制作用。方法人血管内皮细胞株ECV304体外培养,Matrigel胶检测血管内皮细胞的成血管能力,Annexin-Ⅴ/PI双染流式细胞仪检测细胞凋亡,明胶酶谱实验检测血管内皮细胞中MMP-2和MMP-9的活性。结果接种于Matrigel胶的ECV304细胞经β-榄香烯作用后,形成管腔数目明显减少。随着β-榄香烯浓度增加及作用时间延长,ECV304细胞凋亡明显增多。明胶酶谱实验结果显示,β-榄香烯可明显抑制内皮细胞中MMP-2和MMP-9的活性。结论β-榄香烯可以促进体外培养的血管内皮细胞凋亡,抑制血管内皮细胞的成血管能力,使血管内皮细胞在Matrigel胶中形成的管腔数目减少,同时β-榄香烯能够抑制血管内皮细胞中MMP-2和MMP-9的活性。 Objective To investigate the effects of β-demene on the vasculogenesis,apoptosis and the activity of MMP-2,MMP-9 in en- dothelial cells, and to explore the inhibiting effect ofβ-elemene on tumor angiogenesis. Methods ECV304 cells were cultured in vitro, and treated with β-elemene; the angiogenic ability of ECV304 cells was evaluated on Matrigel. Apoptosis were analyzed by using flow cytometry, and the activity of MMP-2 and MMP-9 in ECV304 cells were detected by Zymography. Results Comparing with the control group, the tube formation from the β-elemene treated ECV304 cells on Matrigel notably decreased. With the increase of the dose and administration time, the apoptosis of ECV304 cells was significantly increased, f3-elemene inhibited the activity of MMP-2 and MMP-9 in endothelial cell. Conclusion β-elemene can enhance apoptosis of the endothelial cells cultured in vitro,inhibit their angiogenic ability, reduce the number of tube formation of ECV304 cells on Matrigel, and inhibit the activity of MMP-2 and MMP-9.
