摘要
目的:建立测定连翘中连翘苷含量的超高效液相色谱法。方法:色谱柱为Zorbax Eclipse PlusC1(8100mm×2.1mm,1.8μm),流动相为乙腈-水(24:76,V/V),流速为0.3mL·min-1,检测波长为277nm,柱温为室温。结果:连翘苷的进样量在5.9424~59.4240ng范围内与峰面积积分值呈良好的线性关系(r=0.9993);平均加样回收率为99.83%,RSD=1.27%(n=6)。结论:超高效液相色谱法与传统的高效液相色谱法相比,分析速度快、准确度高、重复性好,减少了有机溶剂的使用,可作为连翘药材质量控制的新方法。
OBJECTIVE: To establish the UPLC method for the determine of phillyrin in Forsythia suspensa. METHODS: The determination was performed on a Zorbax Eclipse Plus C18( 100 mm×2.1 mm, 1.8μm) column with mobile phase consisted of aceto- nitrile-water(24 : 76, V/V) at the flow rate of 0.3 mL .min^-1. The detection wavelength was set at 277 nm and the column tempera- ture was at the room temperature. RESULTS: The linear range was 5.942 4-59.424 0 ng (r= 0.999 3) with an average recovery of 99.83 % (RSD = 1.27 %, n = 6). CONCLUSION: UPLC method is rapid, accurate and reproducible, and it also use the less organic solvent than traditional HPLC method. The method can be used for the quality control of F. suspensa.
