转录因子7样2基因rs290487-T/C多态性与2型糖尿病患者胰岛素抵抗的相关性 被引量:5

Relationship between rs290487-T/C polymorphism of transcription factor 7-like 2 gene and insulin resistance of patients with type 2 diabetes mellitus in Shenyang Region,China

摘要 目的探讨我国沈阳地区汉族人群中转录因子7样2(Transcription factor 7-like 2,TCF7L2)基因rs290487-T/C单核苷酸多态性与2型糖尿病(Type 2 diabetes mellitus,T2DM)患者胰岛素抵抗的相关性。方法选择224例T2DM患者,提取其外周血基因组DNA,采用聚合酶链反应-限制性片段长度多态性技术(PCR-RFLP)对其TCF7L2基因rs290487位点进行基因分型,同时检测各项临床指标,并分析其与TCF7L2基因rs290487-T/C多态性的相关性。结果 T2DM患者组rs290487位点CT+CC基因型的胰岛素抵抗指数(Homeostasis model assessment of insulin resistance,HOMA-IR)、胰岛素β细胞功能指数(Homeostasis model assessment of Beta cell function index,HOMA-β)及空腹胰岛素(Fasting insulin,FINS)均高于TT基因型,差异有统计学意义(P<0.05)。结论沈阳地区汉族人群TCF7L2基因rs290487位点的多态性与T2DM患者胰岛素抵抗相关,为T2DM的病因学研究提供了新的思路。 Objective To investigate the relationship between rs290487-T / C single nucleotide polymorphism of transcription factor 7-like 2(TCF7L2)gene and insulin resistance of patients with type 2 diabetes mellitus(T2DM)in Han population of Shenyang Region,China.Methods Genomic DNAs were extracted from peripheral blood of 224 patients with T2DM,of which the site rs290487 of TCF7L2 gene was genotyped by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP).Various clinical indexes were determined,of which the relationship to rs290487-T / C polymorphism of TCF7L2 gene was analyzed.Results The homeostasis model assessment of insulin resistance(HOMA-IR),function of β cells(HOMA-β)and fasting insulin(FINS) levels of patients whose rs290487 sites were of genotypes CT + CC were significantly higher than those of genotype TT(P 〈 0.05).Conclusion There was a relationship between rs290487-T / C polymorphism of TCF7L2 gene and insulin resistance of patients with T2DM in Han population of Shenyang Region,which provided a novel route for study on etiology of T2DM.
