大孔树脂-梯度高速逆流色谱分离巫山淫羊藿中双藿苷A和朝藿定C 被引量:6

摘要 目的研究大孔树脂结合梯度高速逆流色谱法从巫山淫羊藿粗提物中制备双藿苷A和朝藿定C的分离纯化方法。方法采用大孔树脂法预处理得到巫山淫羊藿粗提物,以二氯乙烷-甲醇-水(4∶4.5∶2,V/V)体系的上相做固定相,下相做流动相,用梯度高速逆流色谱法分离纯化其主要成分双藿苷A和朝藿定C,检测波长为270 nm。结果从1.5 g巫山淫羊藿提取物中分离得到双藿苷A(458 mg)和朝藿定C(321 mg),经HPLC法分析,其纯度分别为95.2%和93.8%。结论该法简单有效,可提高淫羊藿黄酮的分离效率。 AIM To study the method of isolating and purifying epimedin C and diphylloside A simultaneously by macroporous resin-combined with gradient high-speed countercurrent chromatography (HSCCC) from crude ex- tract of Epimedium wushanense T. S. Ying. METHODS Ethylidene chloride-methanol-water (4 : 4. 5 : 2, V/V) was used as biphasic solvent system with a flow rate of 2.0 mL/min. The upper phase was stationary phase while the lower was mobile phase. The wavelength detection was set at 270 rim. RESULTS 458 mg diphylloside A and 321 mg epimedin C were obtained from 1.5 g crude extract of Epimedium wushanense T. S. Ying, and contents were 95.2% and 93.8% , respectively. CONCLUSION The results reveal that the method is high preparative capacity and high efficiency in separation of diphylloside A and epimedin C from Epimedium wushanense T. S. Ying.
