摘要
目的研究Embelin对大肠癌细胞株SW480细胞增殖和凋亡的影响。方法 SW480细胞在一系列浓度Embelin处理48 h以后,噻唑蓝(MTT)法和流式细胞仪分别定量测定细胞增殖抑制率和凋亡率,Western blot检测Cyclin D1、survivin和XIAP的表达水平。结果 Embelin可显著抑制SW480的细胞增殖和诱导凋亡,其作用呈浓度依赖性,作用48 h时半数抑制浓度(IC50)为37.02μM;20μM时细胞凋亡率为(15.3±2.5)%,与40μM时和80μM时细胞凋亡率相比,差异有统计学意义(P<0.05,P<0.01)。但相同浓度的Embelin作用时,细胞的凋亡率低于增殖抑制率。增殖抑制作用可能与下调Cyclin D1蛋白的表达有关。结论 Embelin可显著抑制SW480的细胞增殖和诱导细胞凋亡,并可作为一个潜在的具有抗肿瘤活性的化学药物。
Objective To investigate the effect of Embelin on the cell growth and apoptosis of colorectal cancer cell line SW480.Methods SW480 cells were treated by a series concentration of Embelin.MTT assay and flow cytometry apoptosis assay were used to quantify cell growth inhibition and apoptosis rate respectively.Western blot was used to measure the expression level of Cyclin D1,survivin and XIAP.Results Embelin alone inhibited cell growth and triggered apoptosis of SW480 cells,and IC 50 was 37.02 μM after 48 h.Apoptosis rate was(15.3±2.5)% when treated 20 μM Embelin,and there was significant difference compared with those of 40 μM and 80 μM Embelin(P〈0.05,P〈0.01).But cell apoptosis rate of SW480 was lower than the growth inhibition rate under the same concentration of Embelin.The cell growth inhibition might be associated with the downregulation of cyclin D1.Conclusion Embelin significantly inhibited cell growth and induced apoptosis of SW480 cells,and it may be regarded as a potential anti-cancer drug.
