乙酰肝素酶、蛋白激酶CK2β与缺氧诱导因子-1α在鼻咽癌中的表达与患者预后的关系 被引量:5

摘要 目的探讨乙酰肝素酶(heparanase,H PA)、蛋白激酶C K 2β(protein kinase CK2β,CK2β)、缺氧诱导因子-1α(hypoxia inducible factor-1α,HIF-lα)的表达与鼻咽癌患者预后的关系。方法采用免疫组织化学染色Super-Vision法检测49例鼻咽癌组织标本中HPA、CK2β及HIF-1α的表达水平。结果 Kaplan-Meier单因素分析结果显示,HPA的表达水平与鼻咽癌患者的预后呈显著正相关(Log Rank=11.737,P<0.05),CK2β、HIF-1α的表达水平与鼻咽癌患者的预后亦呈正相关(Log Rank分别为5.538和6.524,P均<0.05)。Cox比例风险模型多因素分析结果显示,仅治疗后有无复发或转移与患者的预后密切相关,HPA、CK2β、HIF-1α的表达均不是患者独立的预后因素。结论 HPA、CK2β和HIF-1α可作为评价鼻咽癌预后的参考指标,具有实用的临床价值。 OBJECTIVE To investigate the expression of HPA,CK2β and HIF-1 α gene in nasopharyngeal carcinoma(NPC)tissues and their correlation with prognosis of NPC.METHODS HPA,CK2β and HIF-1α were detected with Super-Vision immunohistochemical method in 49 NPC specimens.RESULTS In Kaplan-Meier’s univariate analysis,there was a significantly positive correlation between the survival rate and the expression of HPA(Log Rank = 11.7 3 7,P 〈 0.0 5);The survival rates in patients with over expression of CK2β and HIF-1α were significantly lower than those in patients with low expression(Log Rank=5.538 and 6.524,P 〈0.05,respectively).Multivariate Cox regression analysis showed that just whether the recurrence-metastasis group after treatment was independent prognostic factor.None of three genes was independent prognostic factor.CONCLUSION The HPA,CK2β and HIF-1α can be used as a reference indicator of the prognosis of nasopharyngeal carcinoma.
