姜黄素诱导HO-1的表达与抗动脉粥样硬化大鼠的脂质过氧化的研究 被引量:5

Study on the effect of heme oxygenase-1 expression induced by crucumin on anti-lipid peroxidation in the atherosclerotic rats

摘要 目的观察姜黄素诱导HO-1高表达及其对动脉粥样硬化(AS)大鼠血清中氧化低密度脂蛋白(ox-LDL)、丙二醛(MDA)含量的影响。方法将34只健康♂Wistar大鼠,随机分为正常对照组(n=8)、模型组(n=10)、姜黄素组(n=8)及抑制组(n=8),模型组、姜黄素组及抑制组同法复制AS模型,姜黄素组加用姜黄素,抑制组加用姜黄素及锌原卟啉Ⅸ;第14周末处死所有大鼠,取降主动脉检测HO-1的表达、分布及活力,经心脏取血检测ox-LDL、MDA的含量。结果姜黄素组较模型组的HO-1表达量及活力明显增强(P<0.05),ox-LDL、MDA含量显著降低(P<0.05);抑制组较姜黄素组的HO-1表达量及活力明显下降(P<0.05),ox-LDL、MDA含量显著增高(P<0.05)。结论姜黄素可诱导HO-1高表达并显著减轻脂质过氧化程度,锌原卟啉Ⅸ抑制HO-1高表达可使姜黄素抗脂质过氧化作用显著下降。 OBJECTIVE To explore whether curcumin could induce the expression of heme oxygenase-1(HO-1) in rat’s aortic artery and the contents of malondialdehyde(MDA),oxidized low-density lipoprotein(ox-LDL) in atherosclerotic rats.METHODS 34 male Wistar rats were randomly divided into 4 groups as control group(n=8),model group(n=10),curcumin group(n=8) and suppression group(n=8).The rat model with AS was established by the same method in model group,curcumin group and suppression group.Curcumin group was treated with curcumin and suppression group was treated with both curcumin and ZnPP Ⅸ at the same time.All of the rats were executed at the end of 14 weeks.The expression,distribution and activity of HO-1 protein in the aortic artery were measured,and the blood was drawn from heart in each group to evaluate the contents of MDA and ox-LDL.RESULTS The expression and activity of HO-1 in curcumin group were increased significantly than that in model group(P〈0.05),but the contents of MDA and ox-LDL were lower(P〈0.05).The expression and activity of HO-1 in suppression group were decreased significantly than that in curcumin group(P〈0.05),but the contents of MDA and ox-LDL were higher(P〈0.05).CONCLUSION Curcumin could induce HO-1 protein overexpression,increase its activity and decrease the degree of lipid peroxidation.However,this induction was blocked significantly by ZnPP Ⅸ.
