碱性纤维母细胞生长因子对AD细胞模型PKCγ活性影响

Effect of Basic Fibroblast Growth Factor on Protein Kinase Cγ, Activity in PC12 Cells Induced by Aβ25-35

目的 研究碱性纤维母细胞生长因子（bFGF）对β-淀粉样蛋白25-35（Aβ25-35）诱导大鼠肾上腺嗜铬细胞瘤（PC12）细胞存活率、蛋白激酶Cγ（PKCγ）蛋白表达的影响。方法 经体外培养的PC12细胞分正常对照组（常规培养液）、Aβ损伤组（加入培养液及老化Aβ25-35）、bFGF组（加入培养液、bFGF及老化Aβ25-35）。MTT法检测不同处理组PC12细胞存活率，Western Blot免疫印迹法分析PKCγ蛋白表达变化。结果Aβ损伤组PC12细胞存活率低于正常对照组（P〈0．05），bFGF组细胞存活率高于Aβ损伤组（P〈0．05）。Western Blot结果显示，Aβ损伤组PKCγ（0．68±0．07）较对照组PKCγ（0．8±0．08）表达显著下降；Aβ损伤组在加入不同浓度的bFGF后PKCγ的值分别为：1．04±0．10，1．20±0．12，1．39±0．03，1．69±0．16，较Aβ损伤组PKCγ（0．65±0．06）蛋白表达显著增强，并与bFGF浓度呈正相关。结论bFGF对Aβ诱导的PC12细胞损伤具有一定保护作用，可能是通过增强PCI2细胞PKCγ蛋白表达而实现。

Objective To study the effect of basic fibroblast growth factor（bFGF） on cell survive rate and activities of protein kinase C γ（PKC γ） in rat pheochromocytoma（PC12） cells induced by β-amyloid protein（A β 25-35）.Methods Cultural PC12 cells were divided into 3 groups： l.normal control group（added eultrue fluid）;2.A β damaged group（added cultrue fluid and aged A β 25-35）;3.bFGF group（added cultrue fluid,bFGF and aged A β 25-35）.Detected the cell survive rate of every group by MTT, western blot were used to determine PKC γ. Results Cell survive rate of A β damaged group was significantly lower than that in the control（P=0.02）,and cell survive rate of bFGF group was significantly higher than that in the A β damaged group（P=0.01）.Westem blot results demonstrated that the expression of PKC γ （0.68 ± 0.07） in A β damaged group was significantly lower than that in the control （PKC γ ：0.8 ± 0.08）.The expression of PKC γ in different concentration of bFGF group are 1.04 ± 0.10,1.20 ± 0.12,1.39 ± 0.03,1.69 ± 0.16.They were significantly higher than that in the A β damaged group （PKC γ：0.65 ± 0.06）,and had positive correlation with the concentration of bFGF.Conclusion bFGF had protection on PC12 cells induced by A β ,and might procure through enhancing activities of expression of PKC γ.
