二甲氨基偶氮苯(DAB)诱发大白鼠肝癌过程中肝细胞染色质非组蛋白含量和磷酸化作用变化的观察

摘要 对DAB诱癌早期(1~5个月)大白鼠肝细胞核染色质非组蛋白含量和磷酸化作用的变化进行了观察。发现:①诱癌1个月的鼠肝细胞核各部分蛋白质含量和非组蛋白总磷含量没有有意义的变化;②诱癌3个月的鼠肝细胞核各部分蛋白质含量和非组蛋白总磷含量显著增加,鼠肝出现肝硬变的趋势;③诱癌5个月Ⅰ组的鼠肝已形成癌块,非组蛋白含量增加,总磷含量增加,有显著意义。Ⅱ重组鼠出现肝硬化,非组蛋白含量降低,总磷含量比Ⅰ组低。这些结果提示:非组蛋白磷酸化作用与基因表达的调控和癌肿的形成有密切关系。 Nonhistone chromosomal proteins (NHCP) play an important role in regulation of gene expression in eukaryotics. Phosphorylated NHCP stimulates the synthesis of RNA in cell-free system using rat liver RNA polymerase and rat DNA as template have demonstrated by many works recently. Here we described our observation on variance of NHCP and NHCP-phosphorylation using DAB induced hepatoma of rats: (1) Liver muclear protein: DNA and the phosphorylated NHCP showed no significant change at 1 month the DAB diet; (2) Liver muclear proteins: DNA and phosphorylated NHCP showed dignificant increase at 3 months fed DAB diet. meanwhile pathological slices showed pre-carcinoma-tous changes in liver cells; (3) The group Ⅰ of experimental rats that fed DAB diet for 5 months showed different types of changes of hepatoma significantly inreaese in NHCP and phosphorylated NHCP. Another group Ⅱof rats developed cirrhosis with NHCP and phosphorylated NHCP lower than group Ⅰ. Our results suggest that the phosp-liorylation of NHCP is intimately correlated v/ith development of cancer.
