血府逐瘀汤对野百合碱诱导肺血管重构大鼠肺血管平滑肌细胞增殖与凋亡的影响 被引量:10

摘要 目的:观察血府逐瘀汤(XFZYT)对野百合碱(MCT)诱导肺血管重构大鼠肺血管平滑肌细胞增殖与凋亡的干预作用,并探讨其作用机制。方法:雄性SD大鼠54只,随机分为正常对照组、模型组、血府逐瘀汤预防组、血府逐瘀汤治疗组、硝苯地平预防组、硝苯地平治疗组,采用一次性腹腔注射MCT(60mg/kg)复制肺血管重构大鼠模型。药物干预后观察各组大鼠肺血管病理形态学变化,并采用免疫组化技术检测大鼠肺血管增殖细胞核抗原(PCNA)及Caspase-3蛋白表达情况。结果:与模型组比较,各药物组均能使肺小动脉管壁有不同程度变薄、管腔变大(P<0.05~0.01);血管增殖细胞核抗原(PCNA)增殖度下降,Caspase-3蛋白阳性率升高(P<0.05~0.01)。结论:血府逐瘀汤能有效减轻大鼠肺血管重构,其机制可能与调节平滑肌细胞增殖与凋亡有关。 Objective:To observe the intervention effects of xuefu zhuyu decoction on proliferation and apoptosis of pulmonary vascular smooth muscle cells in pulmonary vascular remodeling rats induced by monocrotaline and explore its mechanism.Methods:54 male rats were divided into normal group,model group,xuefu zhuyu decoction prevention and treatment groups,nifedipine prevention and treatment groups,models of pulmonary vascular remodeling were made by single intraperitoneal injecting monocrotaline,each group was given the corresponding treatment,the pathological morphological changes of pulmonary vascular were observed,pulmonary vasculau proliferation cell nuclear antigen(PCNA)and Caspase-3 expression was deternined by immunohistochemisrty methods.Results:The pathological morphological changes of pulmonary vascular in every medicine group were obviously improved at different degree,pulmonary arteriolar wall became thiner,the pulmonary arteriolar lumen got bigger to compare with model group,PCNA epression of pulmonary vascular smooth muscle cells in each medine group was lowered,the positive expression of Caspase-3 raised.Conclusion:Xufu zhuyu decoction could effectively reduce rats' pulmonary vascular remodeling,its mechanism maybe related to regulating proliferation and apoptosis of pulmonary vascular smooth muscle cells.
