摘要
目的测定多囊卵巢综合征(polycystic ovary syndrome,PCOS)患者取卵日卵泡液、血清基质金属蛋白酶(matrix metalloproteinase,MMP)-2、9及基质金属蛋白酶抑制因子(tissue inhibitors of metalloproteinase,TIMP)-1、2的水平,探讨其在PCOS发病中可能的作用机制。方法收集因PCOS接受体外受精-胚胎移植(in vitrofertilization-embryo transfer,IVF-ET)治疗患者30例取卵日卵泡液及血清,以30例同期因输卵管因素接受体外受精(in vitro fertilization,IVF)的卵巢功能正常患者作为对照组,采用酶联免疫吸附测定方法检测卵泡液及血清中MMP-2、9和其抑制物TIMP-2、1的水平。结果①PCOS组卵泡液、血清MMP-2、MMP-9水平均高于对照组(P<0.01),而TIMP-2、TIMP-1水平均低于对照组,但差异无统计学意义(P>0.05);②PCOS组卵泡液、血清MMP-2/TIMP-2、MMP-9/TIMP-1值均高于对照组(P<0.01);③卵泡液、血清MMP-2与TIMP-2水平呈负相关(r=-0.404、-0.448,P<0.05),MMP-9与TIMP-1亦呈负相关(r=-0.428、-0.446,P<0.05)。结论 PCOS患者卵泡液、血清MMP-2,MMP-9水平升高表明,PCOS的多卵泡发育可能与溶解明胶的活性及细胞外基质重塑的异常有关。
Objective To investigate the levels of matrix metalloproteinase (MMP) -2, MMP - 9 and their tissue inhibitors of metalloproteinase ( TIMP - 2 and TIMP - 1, respectively) in the follicular fluid and serum of patients with polycystic ovary syndrome (PCOS) and normally women who receive in vitro fertilization - embryo transfer( IVF - ET) and to determine the MMPs in the pathop PCOS women and 30 possib ovulating le role of hysiology of PCOS. Methods Follicular fluid and serum were collected from 30 normally ovulating women undergoing oocyte retrieval. MMP - 2, MMP - 9, TIMP - 2 and TIMP - 1 were measured by enzyme - linked immunosorbent assay. Results levels of MMP -2 and MMP -9 in follicular fluid and serum were found in the PCOS Significantly higher group compared to the control ( P 〈 0.05 ). TIMP - 2, TIMP - 1 levels in follicular fluid and serum in PCOS group were lower than those in control group, but there were no statistical difference ( P 〉 0.05 ). There were statistical difference between the two groups in MMP - 2/TIMP - 2, MMP - 9/TIMP - 1 in follicular fluid and serum (P 〈 0. 05 ). There were negative correlation between MMP - 2 and TIMP - 2 levels ( r = - 0. 404, - 0.448, P 〈 0.05 ) and MMP - 9 and TIMP - 1 levels ( r = - 0.428, - 0. 446, P 〈 0.05 ) in follicular fluid and serum. Conclusion Significantly higher levels of MMP -2 and MMP -9 in PCOS group may raise the possibility that MMPs may be implicated in the multiple follicular development of PCOS by significantly increased gelatinolytic activity and more intense extracellular matrix remodelling.
