摘要
目的:探讨Ets-1和MMP-9在胎膜早破中的表达和意义。方法:采用免疫组织化学SP法在蛋白水平检测50例胎膜早破者(病例组)及45例无其他产科并发症及内外科合并症的足月择期剖宫产者(对照组)胎膜中Ets-1和MMP-9的表达。结果:①Ets-1蛋白在胎膜滋养层细胞核及胞浆中均有表达,以胞核表达为主;MMP-9主要表达于胎膜滋养层细胞浆中;②Ets-1和MMP-9在胎膜早破组滋养层中高表达,与对照组比较差异有统计学意义(P<0.05);③Ets-1和MMP-9在未足月胎膜早破组滋养层中高表达,但与足月胎膜早破组比较差异无统计学意义(P>0.05)。结论:Ets-1和MMP-9在人类胎膜上均有表达,且在胎膜早破中高表达;Ets-1可能通过上调MMP9的基因表达量使胎膜细胞外基质(ECM)的降解加速,致胎膜紧张度降低而引发胎膜早破。
Objective:To explore the expressions and significances of Ets-1 and matrix metalloproteinase-9(MMP-9) in premature rupture of membrane(PROM).Methods:Immunohistochemical SP method was used to detect the expressions of Ets-1 and MMP-9 in fetal membrane samples of 50 cases with PROM(case group) and 45 full term pregnant women receiving selective cesarean section and without other obstetrical,physical,or surgical complications(control group) at protein level.Results:Ets-1 expressed in cell nucleus and cytoplasm of trophoblasts in fetal membrane,especially in cell nucleus;MMP9 mainly expressed in cytoplasm of trophoblasts in fetal membrane.The expression levels of Ets-1 and MMP-9 in trophoblasts of case group were high,compared with control group,there was statistically significant difference(P0.05).The expression levels of Ets-1 and MMP-9 in trophoblasts of control group were high,compared with case group,there was no statistically significant difference(P0.05).Conclusion:Ets-1 and MMP-9 expressed in human fetal membrane,especially in fetal membrane of cases with PROM;Ets-1 may induce PROM by up-regulating the expression level of MMP-9 gene,accelerating the degradation of extracellular matrix of fetal membrane,and reducing the tension of fetal membrane.
