琼胶酶高产海洋细菌QM42的发酵条件优化 被引量:5

摘要 通过单因素试验和正交试验对琼胶酶高产海洋细菌QM42发酵条件进行了优化。经过发酵培养试验,确定优化培养基组成:蛋白胨浓度5.0g/L、酵母粉浓度0.50g/L、琼胶浓度3.0g/L;在培养初始pH值为7.0、装液量50mL/150mL、培养盐度50g/L,29℃条件下发酵48h,粗酶液酶活力达到627.65U/mL。 The fermentation conditions of an agarase-producing marine bacterium QM42 were optimized by single factor text and orthogonal test.Results showed that the optimal culture medium consisted of 5.0g/L peptone,0.50g/L yeast extract and 3.0g/L agar.The optimal environmental conditions were as followed: temperature 29℃,the initial pH value 7.0,the salinity 50g/L,liquid medium volume 50ml in 150ml shake flask and fermentation time 48h.Under such conditions,agarase activity reached 627.65U/ml.
