摘要
目的:考察厚朴"发汗"对大鼠胃动力的影响和抗溃疡作用。方法:SD大鼠ip左旋精氨酸(L-Arg)5 d建立大鼠胃动力异常模型。胃动力异常大鼠连续5 d分别ig厚朴提取物(厚朴量10 g.kg-1)、"发汗"厚朴提取物("发汗"厚朴量10 g.kg-1),ip西沙必利(0.004 g.kg-1),测定胃残留率,观察厚朴"发汗"前后对胃动力异常大鼠胃动力的改善作用。昆明种小白鼠每只ig 0.35 mL 0.6 mol.L-1盐酸,建立盐酸性溃疡;每只ip吲哚美辛(10 g.kg-1)和ig 0.15 mL 50%乙醇,建立吲哚美辛-乙醇性溃疡模型,连续3 d分别ig厚朴提取物(厚朴量10 g.kg-1)、"发汗"厚朴提取物("发汗"厚朴量10 g.kg-1),盐酸雷尼替丁0.04 g.kg-1;观察厚朴"发汗"前后对小白鼠的抗溃疡作用。结果:大鼠ip L-Arg造成大鼠胃动力减缓、血浆胃动素减少。厚朴、"发汗"厚朴提取液均能有效抑制L-Arg引起的大鼠胃残留率和胃动素的下降(P<0.05),"发汗"厚朴组与厚朴组、西沙必利组之间比较有显著性差异(P<0.05)。厚朴组、"发汗"厚朴组、西沙必利组的胃残留率分别为(70.2±14.1)%,(61.5±12.8)%,(71.3±12.4)%;胃动素分别为(84.8±11.6),(92.9±11.7),(85.4±12.1)nmol.L-1。厚朴、"发汗"厚朴均能减小盐酸性溃疡指数、吲哚美辛-乙醇性溃疡指数(P<0.01),"发汗"厚朴组盐酸性溃疡指数、吲哚美辛-乙醇性溃疡指数要小(P<0.01)。厚朴组、"发汗"厚朴组、盐酸雷尼替丁组盐酸性溃疡指数分别为(36.8±10.4),(29.5±8.2),(17.5±10.1)mm;吲哚美辛-乙醇性溃疡指数分别为(45.4±11.7),(33.8±11.0),(20.3±4.9)mm。结论:厚朴"发汗"后改善大鼠胃动力障碍和抗溃疡作用更强。厚朴"发汗"炮制工艺是必要的。
Objective: The influences of Magnoliae Officinalis Cortex on stomach dynamic and ulcer in ratsbefore and after its diaphoretic processing were investigated. Method: The gastric motility disorder in SD rats was established by injecting L-Arg intraperitoneally for five days, then the rats were given the extract of Magnoliae (Magnoliae Officinalis Cortex 10 g·kg^-1) by intragastric administration, the extract of diaphoretic Magnoliae (diaphoretic Magnoliae Officinalis Cortex 10 g·kg^-1) was gived by intragastric administration, cisapride (0. 004 g·kg^-1) by intraperitoneal injection, respectively, gastric residual rate was determined, then improving effect by Magnolia Officinalis Cortex before and after its diaphoretic processing was observed. Kunming mice were given 0.35 mL 0.6 mol·L^-1 hydrochloric acid by ig, HCI-induced gastric ulcer was established, mice were given 10 g·kg^-1indomethacin by ip and 0. 15 mL 50% ethanol by ig, indomethacin-ethanol-ulcer was established, and antiulcer effect between pre-and post treatment of diaphoretic Magnoliae Officinalis Cortex was observed by giving the extract of Magnoliae Officinalis Cortex (Magnoliae Officinalis Cortex 10 g·kg^-l), the extract of diaphoretic Magnoliae Officinalis Cortex (diaphoretic Magnoliae Officinalis Cortex 10 g·kg^-1), 0. 04 g· kg^-1 ranitidine hydrochloride for continuous 3 d. Result: Gastrointestinal motility in rats injected by L-Arg intraperitoneally was slowed down, plasma motilin was reduced. The extract of Magnoliae Officinalis Cortex, the extract of diaphoretic Magnoliae Officinalis Cortex could effectively inhibit the decrease in Gastrointestinal motility (P 〈 0. 05 ) and plasma motilin (P 〈 0.05) , and there was a significant difference between the diaphoretic Magnoliae Officinalis Cortex group and the Magnoliae Officinalis Cortex group, and the diaphoretic Magnoliae Officinalis Cortex groupand the group cisapride (P 〈 0.05). Gastric residual rate in the extract of Magnoliae Officinalis Cortexand the extract of diaphoretic Magnoliae Officinalis Cortex group, cisapride was (70.2±14. 1 ) % (61.5±12.8)%, (71.3±12.4)% respectively; plasma motilin was (84.8±11.6), (92.9±11.7), (85.4±12.1) nmol·L^-1. Both Magnoliae Officinalis Cortex and diaphoretic Magnoliae Officinalis Cortex could reduce the index of HCl-ulcer and indomethacin-ethanol-ulcer (P 〈 0.01 ) , and the index of HCl-ulcer and indomethacin-ethanol-ulcer of diaphoretic Magnoliae Officinalis Cortex was smaller to that of Magnoliae Officinalis Cortex (P 〈 0.01 ). The index of HC1- ulcer in the extract of Magnoliae Officinalis Cortex, the extract of diaphoretic Magnoliae Officinalis Cortex and ranitidine hydrochloride groups was (36.8±10.4), (29.5± 8.2), (17.5±10. 1) mm respectively; indomethacin-ethanol-ulcer were (45.4 ±11.7) , (33.8±11.0), (20.3±4.9) mm. Conclusion: Diaphoretic Magnoliae Officinalis Cortex is better than Magnoliae Officinalis Cortex on improving the gastric motility disorder and antiulcer effect, so processing technology of diaphoretic Magnolia Officinalis Cortex is necessary.
