摘要
目的研究参葛降糖胶囊对胰岛素抵抗糖尿病大鼠脂肪细胞因子水平的影响,探讨参葛降糖胶囊治疗糖尿病的作用机制。方法采用灌胃高脂高糖脂肪乳加腹腔注射小剂量链脲佐菌素(STZ)的方法建立胰岛素抵抗糖尿病大鼠模型。造模成功后随机分为正常对照组、模型组、阳性对照组和参葛降糖胶囊低、中、高剂量组,分别给予相应的干预,共30天。检测各组大鼠总胆固醇(TC)、三酰甘油(TG)、游离脂肪酸(FFA)、瘦素(leptin)和肿瘤坏死因子-α(TNF-α)水平。结果与正常对照组比较,参葛降糖胶囊中、高剂量组TC、TG、TNF-α、leptin含量差异无统计学意义(P>0.05),而FFA含量差异有统计学意义(P<0.001,P<0.01);参葛降糖胶囊低剂量组大鼠血清TC、TG、TNF-α、FFA含量差异有统计学意义(P<0.001,P<0.01,P<0.05),而leptin水平差异无统计学意义(P>0.05)。与模型组比较,参葛降糖胶囊高、中剂量组大鼠血清FFA、TC、TG、leptin、TNF-α含量降低显著,差异有统计学意义(P<0.001,P<0.01);参葛降糖胶囊低剂量组大鼠血清TC、TG、leptin含量降低,差异有统计学意义(P<0.001,P<0.01,P<0.05),而FFA、TNF-α含量差异无统计学意义(P>0.05)。结论参葛降糖胶囊可以通过影响胰岛素抵抗糖尿病大鼠脂肪细胞因子水平,调节脂代谢,改善胰岛素抵抗。
Objective To observe the effect of "Shen' ge Jiangtang Capsule" on adipose cytokine levels in rats with insulin resistance diabetes in an attempt to explore its mechanisms. Methods The models were established by feeding high glucose and lipoids food and injecting small dose of streptozotocin. Then the models were randomized into normal control group, model group, positive control group and "Shen' ge Jiangtang Capsule" groups (small, medium and large dose) , each group was administered with gastric lavage for 30 days. The TC, TG, FFA, leptin and TNF-α were detected. Results Compared with the normal control group, no significantly difference was found in the levels of TC, TG, TNF-α and leptin between medium and large dose group ( P 〉 0.05 ), but there, was a significant difference in FFA level (P 〈 0. 001, P 〈 0. 01 ) ; the levels of TC, TG, TNF-α and FFA in small dose group showed significant differences ( P 〈 0. 001, P 〈 0.01, P 〈 0.05 ) , but no significant difference was found in leptin level (P 〉 0.05 ). Compared with model group, the levels of TC, TG, FFA, leptin and TNF- α in medium and large dose groups were decreased significantly ( P 〈 0. 001, P 〈 0.01 ) ; the levels of TC, TG and leptin in small dose were decreased significantly (P〈O. 001, P〈0. 01 ,P〈0.05) , but no significant difference was found in the levels of FFA and TNF-α (P 〉0.05). Conclusion "Shen' ge Jiangtang Capsule" can regulate lipid metabolism and improve insulin resistance by influencing the adipose cytokine levels of rats with insulin resistance diabetes.
