摘要
目的观察联合缺氧诱导因子-1α(HIF-1α)和bcl—xL小干扰RNA对肺腺癌A549细胞增殖的影响。方法体外培养的A549细胞分为4组:A组用生理盐水处理,B组用HIF-1α小干扰RNA处理,C组用bcl—xL小干扰RNA处理,D组用HIF-1α和bcl—xL小干扰RNA联合处理。逆转录一聚合酶链反应(RT—PCR)检测HIF-1α和bcl-xL的mRNA表达,Western—blot检测两种蛋白的表达,噻唑蓝(MTT)比色法检测A549细胞增殖。结果B组和D组细胞的HIF-1α mRNA表达量分别下降至0.733±0.157、0.766±0.208,蛋白表达量下降至0.372±0.095、0.395±0.077,与A、C组比较差异有统计学意义(P〈0.05);C组和D细胞的bcl—xLmRNA表达量分别下降至0.514±0.183、0.509±O.168,蛋白表达量下降至0.447±0.097、0.410±0.089,与A、B组比较差异有统计学意义(P〈0.05);D组的细胞增殖抑制最明显,与对照组比较差异有统计学意义(P〈0.05)。结论HIF-1α和bcl-xL小干扰RNA联合应用能够明显抑制A549细胞增殖。
Objective To explore the effect of hypoxia inducible factor 1 alpha (HIF-lct) and bcl- xL small interfering RNA in combination on the proliferation of lung adenocarcinoma A549 cells. Methods A549 cells in vitro were divided into four groups: group A treated with normal saline, group B with HIF-1α siRNA, group C with bel-xL siRNA, group D with HIF-1α siRNA and bcl-xL siRNA. The expression of HIF-1α and bcl-xL mRNA was detected by using reverse transeription-polymerase chain reaction (RT-PCR), their proteins were detected by using Western blotting, and MTT assay was used to detect the growth of A549 cells. Results In groups B and D, the expression level of HIF-1α mRNA was decreased to 0. 733 ± 0. 157 and O. 766 ±0. 208, and its protein was decreased to 0. 372±0. 095 and 0. 395±0. 077, significantly lower than in groups A and C (P 〈 0. 05 ). In groups C and D, the level of bcl-xL mRNA was decreased to 0. 514±0. 183 and 0. 509 ±0. 168, and its protein was decreased to 0. 447±0. 097 and 0. 410 ±0. 089, sig- nificantly lower than than in groups A and B (P 〈 0. 05 ). The growth of A549 cells in group D was inhabited strongly, and there was significant difference between group D with groups A, B and C (P 〈 0. 05). Conclu- sion HIF-1α siRNA combined with bcl-xL siRNA can significantly suppress the growth of 549 ceils.
