摘要
目的:观察小蔓长春花提取物(vinca minor linn extraction,VMLE)对实验性脑缺血大鼠血液及抗氧化能力的影响。方法:根据Zea-Longa线栓法制作SD大鼠右侧大脑中动脉阻塞(middle cerebral arteryocclusion,MCAO)模型,清醒后1h,即参考Bederson方法对模型大鼠进行神经功能缺损评分,入选大鼠被随机分为模型组(model)、尼莫地平组(nimodipine)、VMLE高、低剂量组4组,另加假手术组(sham)共5组,各组遂即分别给予相应药物灌胃2d。48h后测定大鼠血液流变学和抗氧化能力。结果:VMLE10g/kg、5g/kg均能明显降低,实验性脑缺血大鼠全血黏度、血浆黏度、红细胞压积(Hct)、红细胞聚集指数(EAI)、红细胞刚性指数(ERI);增强了SOD和GSH-PX活性,降低了MDA含量,有统计学意义(P<0.05);尼莫地平10g/kg能增强SOD和GSH-PX活性,降低了MDA含量,但对实验性脑缺血大鼠全血黏度、血浆黏度、红细胞压积无显著影响。结论:VMLE可以改善脑缺血大鼠血液流变学及抗氧化能力。
Objective To study the effects of Vinca minor linn extraction (VMLE) on the hemorheology and antioxidant capacity in rats with experimental cerebral ischemia. Methods Rat models with middle cerebral artery occlusion (MCAO) were made by Zea-Longa traditional method. Then the scores of behavior were evaluated in MCAO rats hy Bederson method. All the rat models were randomly divided into 4 groups : model group, nimodipine group, VMLE high and low dose groups (10 g/kg and of 5 g/kg of VMLE ), and sham group. Rats in each group received corresponding drugs by gavage for 2 days. The hemorheology and antioxidant capacity of rats were detected 48 hours later. Results Blood viscosity, plasma viscosity, hematocrit, erythrocyte aggregation index, erythrocyte rigidity index, and level of MI)A were significantly decreased and SOD and GSH-PX activity were enhanced in VMLE high and low dose groups (P 〈 0.05) ; Nimodipine could enhance SOD and GSH-PX activity and decrease the level of MDA, but couldn't affect the blood viscosity, plasma viscosity, and hematocrit. Conclusion VMLE can significantly improve hemorheology and antioxidant capacity in rats with cerebral ischemia.
