细胞间黏附分子-1在大鼠急性坏死性胰腺炎相关性肺损伤中的表达和意义 被引量:2

摘要 目的:探讨细胞间黏附分子在大鼠急性坏死性胰腺炎相关性肺损伤中的表达和意义。方法:80只雄性Sprague-Dawley大鼠随机分为两组,假手术组(SHAM)和胰腺炎肺损伤组(ALI)。术后6、12、24、48h分别处死大鼠观察胰腺,肺脏病理学形态改变;检测血清淀粉酶、TNF-α、IL-6含量;称肺干湿重比;检测肺组织髓过氧化物酶(MPO)活性;荧光定量PCR检测肺组织细胞间黏附分子-1(ICAM-1)mRNA表达;免疫印迹法检测肺组织中ICAM-1蛋白表达。结果:ALI组各时点肺湿干质量比较SHAM组有显著差异(P<0.05),MPO活性测定各时点均较SHAM组升高(P<0.05);ALI组血清淀粉酶、TNF-α、IL-6含量均较SHAM升高(P<0.05);ALI组ICAM-1mRNA表达较SHAM组明显升高,并且随时间进展而逐渐增加,12h达高峰,并维持在高水平表达;同时ICAM蛋白表达也逐渐升高,12h达高峰后持续高水平表达;TNF-α与ICAM-1mRNA及蛋白质表达变化呈正相关趋势。结论:在大鼠急性坏死性胰腺炎肺组织中,ICAM-1在基因及蛋白水平上呈过度表达,并且与肺组织损伤程度密切相关,前炎性因子TNF-α在ICAM-1的过度表达中起到了重要作用。
