摘要
目的探讨血管内皮生长因子及其受体在血管壁外膜成纤维细胞的表达。方法采用ELISA方法来检测外膜成纤维细胞血管内皮生长因子的分泌及TGF-β、bFGF、AngⅡ、PDGF对血管内皮生长因子分泌的影响,使用Western blot法观察TGF-β、bFGF、AngⅡ、PDGF等对血管内皮生长因子I型受体表达的影响。结果血管外膜成纤维细胞能够分泌血管内皮生长因子。TGF-β能显著增高血管内皮生长因子分泌(n=5,P<0.05),而PDGF、bFGF、AngⅡ对血管内皮生长因子分泌比较没有统计学差异(n=5,P>0.05)。TGF-β、PDGF、bFGF、AngⅡ均可刺激外膜成纤维细血管内皮生长因子I型受体表达增高(n=4,P<0.05)。结论血管壁外膜成纤维细胞可以分泌血管内皮生长因子,血管局部活性物质能有效刺激其受体表达增高。因此推测血管内皮生长因子可能介导血管外膜成纤维细胞参与血管壁的重构。
Objective To explore the expression of VEGF and its receptors in adventitial fibroblasts. Methods Aortic adventitial fibroblasts from Sprague-Dawley (SD) rats were cultured. VEGF were measured in AFs by enzyme-linked immunoabsorbent assay (ELISA), and TGF-β, bFGF, PDGF, AngII on the VEGF secretion, the effect of TGF-β, bFGF, PDGF, Ang II on the expression of VEGFR-1 was detected by western blot. Results Secretion of VEGF by adventitial fibroblasts. ELISA results showed that the VEGF protein was expressed in cultured serum-free medium (SFM) in time dependent manner(n=4, F=59.538,P=0.000), and TGF-β can significantly increase the VEGF expression. It suggested that VEGF can be secreted from fibroblasts into conditional medium.Western blot results showed that VEGFR-1 was expressed in fibroblasts. TGF-β(n=4,P〈0.01), bFGF, PDGF, Ang II (n=4,P〈0.05) can increase the VEGFR-1 expression. Conclusion Our results demonstrated the existence of VEGF and its receptor VEGFR-1 in adventitial fibroblasts. This might be an important mediator in the pathogenesis of vascular remodeling.
