RO3306对人结肠癌细胞HT29增殖及凋亡的影响

摘要 目的观察RO3306对人结肠癌细胞HT29增殖及凋亡的影响。方法选用苔盼蓝染色细胞计数法、细胞集落形成法检测RO3306对人结肠癌细胞HT29细胞增殖影响;采用流式细胞术检测RO3306对HT29细胞周期及凋亡指数的影响。结果苔盼蓝染色和集落形成法结果均显示,RO3306作用HT29细胞后,细胞的生长抑制率随着药物浓度的增加、作用时间的延长而逐渐增高;流式细胞仪检测结果显示,RO3306在0.2mg/L,2mg/L浓度下作用48h对HT2凋亡指数呈现随浓度的提高而逐步上升的趋势,当药物浓度增加到20mg/L,HT29的凋亡指数显著上升,其细胞周期有明显阻滞,随着作用时间的推移出现了更明显的S期阻滞。结论 RO3306具有时间和浓度依赖性地抑制大肠癌HT29细胞的增殖,阻滞其细胞周期、诱导其凋亡。
