摘要 目的探讨姜黄素衍生物FM04体外抗淋菌的效果与机制。方法应用K-B法检测40株淋菌对抗菌药物的敏感性,煮沸法提取菌株DNA,随机抽取增其中22株PCR扩TEM、tetM、erm和mefA基因;采用滤纸片扩散法,初步判断FM04是否有抗菌作用,作用结果结合耐药表型和基因型分析FM04体外抗淋菌作用可能的机制。结果淋菌的敏感性测定显示多药耐药现象明显;TEM、tetM的检出率均为77.3%,未检出erm和mefA基因;FM04对8株淋菌的抑菌环直径均为10~13mm,差异无统计学意义。结论姜黄素衍生物FM04具有体外抗淋菌作用,有进一步开发研究的前景;未发现其作用机制与淋菌耐药表型及基因型有关系。 OBJECTIVE To analyze the effect and mechanism of in vitro anti-gonococcus activity of curcumin derivative FM04. METHODS The drug susceptibility of 40 strains of Neissena gonorrhoeae was tested by K-B and agar dilution methods. Total DNA of those strains was extracted by boiling method, 22 strains of N. gonorrhoeae were randomly selected for the amplification of the resistance genes of TEM, terM, erm and mefA by PCR; by the means of filter paper diffusion, preliminary judgment of the antibacterial activity of FM04 was performed, the mechanism of in vitro anti-gonococcus activity of FMO4 was analyzed through the combination with the effect result and drug resistant phenotype. RESULTS The susceptibility testing showed that the multidrug-resistance of N. gonorrhoeae was significant; the detection rates of both TEM and tetM were 77.3%, respectively. The genes of erm and rnefA were not detected; the bacteriostatie annulus diameter of FM04 to 8 strains of N. gonorrhoeae were 10- 13 mm, the difference was not statistically significant. CONCLUSION FM04 exhibits the in vitro anti-gonococcus activity, which deserves further research and development; there is no correlation between the mechanism of FM04 and the drug resistant phenotype and genotype of N. gonorrhoeae.
