耐力运动锻炼对正常及肺气肿大鼠膈肌、腓肠肌和心肌氧化性能的影响 被引量:4

Effects of Endurance Training on Oxidative Capacity of Diaphragm,Gastrocnemius and Heart Muscle in Normal and Emphysematous Rats

摘要 目的 :观察中等强度 (5 5 %~ 75 %V0 2max)耐力运动锻炼对正常及肺气肿大鼠膈肌、腓肠肌和心肌氧化性能的影响。方法 :SD大鼠先随机分为 2组 ;肺气肿组经气管内一次性缓慢滴注猪胰弹性蛋白酶 80 0 EU/kg ,正常组一次性气管内滴注等体积 0 9%氯化钠。 4周后分别将 2组大鼠再随机分为静息对照和运动组共 4组 ,即 :正常对照组 (NC ,n=10 ) ,正常运动组 (NE ,n =10 ) ,肺气肿对照组 (EC ,n =12 )和肺气肿运动组 (EE ,n =11)。然后NE组和EE组进行同等强度和时间的电动平板跑步锻炼。在末次运动锻炼结束后 48~ 72小时处死大鼠 ,用组织化学方法染色 ,光密度分析测定膈肌肋部、腓肠肌和心肌纤维琥珀酸脱氢酶 (SDH)和乳酸脱氢酶 (LDH)活性 ,评价相应肌组织的氧化代谢能力。结果 :运动锻炼后NE和EE组心肌和腓肠肌肌纤维氧化性能明显改善 ,SDH活性升高 (P <0 0 5 ) ,但膈肌肌纤维中SDH活性仅在EE组有提高 (P <0 0 5 )。LDH在各组不同肌纤维中均无明显变化。 To investigate the effects of moderate intensity endurance training (55%—75%VO 2max )on biochemical responses of diphragm,gastrocnemius and heart in normal and emphysematous rats, SD rats were randomly divided into two groups first.One was emphysematous model by single endotracheal instillation of porcine pancreatic elastase 800 EU /Kg,the other was normal control received single endotracheal instillation of equal volume 0.9% NaCl.Following a 4-week recovery period,each group was randomly divided into sedentary and exercise endurance training subgroups.They were normal control(NC,n=10),normal exercise (NE,n=10),emphysematous control (EC,n=12)and emphysematous exercise (EE,n=11).The NE and EE groups were subjected to a progressive training on motor driven treadmill,5 days a week for 12 weeks. After 48~72h of the end of exercise training,the biochemical activity of succinate dehydrogenase (SDH) and lactate dehydrogenase (LDH) in the muscle fibers of costal diaphragm,gastrocnemius and heart were determined.The results showed that endurance training enhanced oxidative capacity in the heart and gastrocnemius of NE and EE groups(p<0 05),and in the costal diaphragm of EE group (p<0 05),as evidenced by SDH as well.Furthermore,training did not alter LDH activity in the costal diaphragm,gastrocnemius and heart muscle fibers in all groups.
