腺病毒介导的人GM-CSF基因转染瘤苗增强机体免疫功能的实验研究 被引量:1

摘要 目的 研究腺病毒介导的人 GM- CSF基因转染瘤苗体内抗肿瘤免疫作用极其机理。方法 应用 GM- CSF基因转染、辐照的瘤苗对小鼠肝癌模型进行免疫基因治疗 ,观察该疗法对荷瘤小鼠脾细胞 NK、L AK、CTL 活性的影响、脾淋巴细胞转化反应的影响以及荷瘤小鼠的生存期。结果 经 GM- CSF基因转染瘤苗治疗后 ,脾细胞 CTL活性显著升高 ,而 NK、L AK细胞活性未见明显增强、脾淋巴细胞转化反应显著增强、生存期显著延长。结论  GM- CSF基因转染瘤苗能诱导出有效的抗肿瘤免疫反应 ,揭示了应用该疗法进行肿瘤基因治疗的可行性。 Objective To study the antitumor effect of cancer vaccine transfected with human GM CSF gene in vivo and its mechanism. Methods The NK, LAK, CTL activity and the lymphocyte transformation test (LTT) of splenocytes from tumor bearing mice after treated with GM CSF transfected and irradiated cancer cells were measured. The survival time was also observed. Results Treated with GM CSF transfected and irradiated cancer cells not only significantly augmented LTT and CTL activity (but not NK or LAK activity) of splenocytes, and also significantly prolonged the survival time of tumor bearing mice.Conclusion Irradiated and GM CSF transfected tumor cells induced effective anti tumor immunity. The expression of GM CSF gene in tumor cells seems to be feasible for cancer gene therapy.$$$$
