扶正消瘤汤抑制人肝癌细胞SMMC-7721的增殖和诱导凋亡的实验研究

Experimental research of Fuzheng xiaoliu decoction on inhibiting proliferation and inducting apoptosis of human hepatoma SMMC-7721 cells

目的探讨扶正消瘤汤对人肝癌细胞SMMC-7721的增殖和诱导凋亡的作用。方法 MTT法测定扶正消瘤汤对人肝癌细胞SMMC-7721的增殖的影响;Elisa法测定肿瘤坏死因子(TNF-)生成;流式细胞法检测扶正消瘤汤对人肝癌细胞SMMC-7721凋亡的影响;免疫组化法测定bcl-2和bax表达。结果扶正消瘤汤100 g/ml、50 g/ml剂量组在12～84 h对人肝癌细胞SMMC-7721的生长有明显的抑制作用,24～48 h对SMMC-7721细胞TNF-生成均有明显的促进作用,对SMMC-7721细胞凋亡具有明显的促进作用,并随浓度和时间(24～72 h)的增加凋亡率增加,活细胞减少;扶正消瘤汤高剂量组对SMMC-7721细胞bax表达有明显上调作用,浓度越大上调越明显。扶正消瘤汤高、低剂量组对SMMC-7721细胞bcl-2表达有明显下调作用,其中高剂量组与模型组差异有统计学意义(<0.05)。结论扶正消瘤汤对肿瘤的治疗作用的机制可能与促进TNF-生成有关,并且与抑制肿瘤细胞增殖、诱导肿瘤细胞凋亡、调控bax、bcl-2表达有关。

Objective To investigate the effect of Fuzheng xiaoliu decoction on inhibiting proliferation and induc- ting apoptosis of human hepatoma SMMC-7721 cells. Methods The influence of Fuzheng xiaoliu decoction on the proliferation of human hepatoma SMMC-7721 cells was determined by MTT assay. TNF-ct production was de- termined by Elisa. The influence of Fuzheng xiaoliu decoction on apoptosis in human hepatoma SMMC-7721 cells was detected by flow cytometry. Immune staining method was used to determine bcl-2 and bax expressions.Results The results showed 100 μg/ml and 50 μg/ml dose groups could significantly inhibit the growth of human hepatoma SMMC-7721 cells from 12 hours to 84 hours. It could obviously promote tumor necrosis factor-α （TNF-α） generation of SMMC-7721 cells from 24 hours to 48 hours and apoptosis of SMMC-7721 cells, and the apoptosis rate increased with the concentration and time （24 - 72 h）. High dose group had significant up- regulation effect of SMMC -7721 cells bax expression, and the effect was more obvious with the higher concentration. Both high and low-dose groups could significantly down-regulate bcl-2 expressions, and there was a statistical significance between high-dose group and model group（P 〈 0.05）. Conelusious The mechanism ofFuzheng xiaoliu decoction on tumor maybe related with promotingTNF-ct generation, inhibiting tumor cell proliferation, inducing tumor cell apoptosis and regulating bax, bel-2 expressions.
