PCR扫描技术筛选新型活性微生物代谢产物的研究进展

Progress of screening new active microbial metabolites by PCR scanning technology

摘要 PCR扫描筛选技术以保守序列为基础设计引物,以基因文库为模板进行扩增,获得新型活性微生物代谢产物的基因,继而通过基因重组技术获得对应活性代谢产物,克服了传统筛选方法必须获得微生物纯培养物的限制,有效提高新型活性微生物代谢产物发现的概率。本文综述利用PCR扫描技术筛选新型活性微生物代谢产物的基本方法及其在聚酮类化合物和新型酶类筛选中的应用进展。 PCR scanning technology is based on the conserved sequences to design a primer from gene library as template to amplify the gene of new active microbial metabolites and then to get the products by gene recombination. This new technology can overcome the disadvantage of the traditional method which has to get the microbial pure culture, and can increase the probability of getting new active microbial metabolites. The basic method, application progress of this technology and the outlook of application prospect are summarized in this review.
