摘要
目的观察人参皂苷Rh2对人肝癌Bel-7402细胞增殖和凋亡的影响,并初步研究其机制。方法将Bel-7402细胞与人参皂苷Rh(2200、100和50μg/mL)共同培养48 h。MTT法检测人参皂苷Rh2对Bel-7402细胞增殖的影响;流式细胞仪分析细胞周期中各时期的细胞百分数;TUNEL技术检测其对Bel-7402细胞凋亡的影响;Western blotting法检测Bel-7402细胞中Bcl-2、Bax的蛋白表达。结果 MTT结果显示,人参皂苷Rh2能抑制Bel-7402细胞的生长;流式细胞检测显示,人参皂苷Rh2能使细胞停滞于G1期;TUNEL结果显示,人参皂苷Rh2能有效诱导Bel-7402细胞凋亡;Western blotting结果显示,人参皂苷Rh2能上调Bel-7402细胞中Bax的表达,下调Bcl-2的表达。上述结果均在一定程度上呈量效关系。结论人参皂苷Rh2能抑制Bel-7402细胞生长并促进其凋亡,其作用机制可能为上调Bax和下调Bcl-2。
[Objective] To investigate the effects of ginsenoside Rh2 on the proliferation and apoptosis of human liver cancer cells Bel-7402. [Methods ] Human hepatocarcinoma Bel-7402 cells were co-cultured with gensenoside Rh2 (200, 100, 50 μg/mL) for 48 h. MTT method was performed to detect the effect of gensenoside Rh2 on Bel-7402 cells proliferation. FCM was used to analyze the ratio of Bel-7402 cells in each phase of cell cycle. TUNEL technique was performed to analyze the effect of gensenoside Rh2 on Bel- 7402 cells apoptosis. Western blotting was used to detect the protein expression of Bcl-2 and Bax in Bel- 7402 cells. [Results ] MTT showed that the gensenoside Rh2 could inhibit the proliferation of Bel-7402 cells. FACS showed that the gensenoside Rh2 could retard Bel-7402 cells in G1 phase of cell cycle. TUNEL showed that the gensenoside Rh2 could promote Be1-7402 cells apoptosis. Western blotting showed that in Bel-7402 cells, the expression of Bcl-2 was decreased and the expression of Bax was increased. Each result above showed a dose-response relationship to a certain degree. [Conclusion] The gensenoside Rh2 could inhibit the proliferation as well as promote the apoptosis of Bel-7402 cells, the mechanism of which may involve the down-regulation of Bcl-2 and the up-regulation of Bax.
