摘要
目的:建立HPLC同时测定栀子中8个环烯醚萜苷类成分的含量测定方法。方法:采用Kromasil C18色谱柱(4.6mm×250 mm,5μm),流动相为乙腈-水-三氟乙酸(6∶94∶0.05),流速1.0 mL.min-1,检测波长238 nm,柱温40℃。结果:栀子苷,栀子新苷,山栀苷,栀子苷酸,去乙酰车叶草苷酸甲酯,羟异栀子苷,鸡矢藤次苷甲酯和京尼平龙胆二糖苷的线性范围(μg)和加样回收率分别为1.503 6~15.036(99.6%),0.042 56~0.425 6(100.6%),0.103 8~1.038(101.2%),0.009 92~0.099 2(99.5%),0.023 32~0.233 2(100.3%),0.412 8~4.128(98.7%),0.020 40~0.204 0(99.8%)和0.465 6~4.656(100.1%)。结论:该方法分离良好,简便准确,重复性好,可更好地为栀子药材的质量控制提供依据。
Objective: To establish an HPLC method for simultaneous determination of eight iridiods in Gardeniae Fructus. Method: Kromasil C ls column (4. 6 mm x 250 mm, 5 μm) was adopted, with acetonitrile-water-trifluoroacetic acid (6: 94: 0. 05) as the mobile phase at the flow rate of 1.0 mL . min - 1. The detection wavelength was set at 238 nm, and the column temperature was 40 °C. Result: The linear ranges of geniposide, gardoside, shanzhiside, geniposidic acid, deacetyl asperulosidic acid methyl ester, gardenoside, scandoside methyl ester, and genipin gentiobioside were 1. 503 6-15.03 6, O. 042 56-0. 425 6, 0. 103 8-1. 038, 0. 009 92- 0. 099 2, 0. 023 32-0. 233 2, 0. 412 8-4. 128, 0. 020 40-0. 204 0 and 0. 465 6-4. 656 μg, respectively. Their average recoveries were 99. 6%, 100. 6%, 101.2% , 99. 5%, 100. 3%, 98.7%, 99. 8% and 100. 1%, respectively. Conclusion: The method shows good separation and it is so simple, accurate and highly repeatable that it can be used for providing basis for quality control of Gardeniae Fructus.
