双靶点沉默Survivin和RhoA对肺腺癌A549细胞的作用

摘要 目的双靶点靶向沉默肺腺癌A549细胞Survivin和RhoA基因表达,研究其治疗作用。方法构建共表达靶向Survivin和RhoA基因的串联microRNA干扰载体,脂质体介导该干扰载体转染A549细胞作为实验组,空白干扰载体作为对照组。于转染48 h后,RT-PCR检测Survivin、RhoA mRNA表达,Western blot检测Sur-vivin、RhoA蛋白质表达,MTT法检测细胞的增殖并计算抑制率,流式细胞仪检测细胞凋亡。结果实验组SurvivinmRNA表达抑制率为62.7%,RhoA mRNA表达抑制率为69.0%;实验组Survivin蛋白表达抑制率为66.7%,RhoA蛋白表达抑制率为71.9%。与对照组比较,实验组细胞增殖抑制率为(47.63±6.34)%。实验组细胞凋亡率为(36.32±6.42)%,明显高于对照组[(0.62±0.2)%](P<0.05)。结论成功构建Survivin-RhoA串联microR-NA干扰载体,表达的人工双靶点microRNA能通过促进细胞凋亡来抑制A549细胞增长。
