摘要
目的探讨冠状动脉粥样硬化患者外周血蛋白激酶C受体蛋白(RACK1)的表达变化及与冠状动脉狭窄程度的关系。方法入选初诊为冠状动脉粥样硬化性心脏病的患者83例,根据冠状动脉造影结果,采用冠状动脉狭窄程度积分评价其狭窄程度。患者被分为4组:Ⅰ组(0分)即为对照组;Ⅱ组(1~10分);Ⅲ组(11~20分);Ⅳ组(>20分)。采用实时荧光定量聚合酶链反应(real-time PCR)检测所有患者外周血RACK1 mRNA的表达情况。结果Ⅱ、Ⅲ和Ⅳ组的RACK1 mRNA表达水平均显著高于Ⅰ组(t分别为10.62,12.90,14.00,均为P<0.05),而Ⅲ组和Ⅳ组又显著高于Ⅱ组(t分别为9.40和10.74,均为P<0.05),Ⅲ组、Ⅳ组两组间比较差异无统计学意义(t=1.43,P>0.05)。相关分析显示,冠状动脉粥样硬化患者外周血RACK1 mRNA表达水平与冠状动脉狭窄程度存在相关性,相关系数rs=0.59(P<0.01)。结论 RACK1在冠状动脉粥样硬化患者外周血中表达上调,且与冠状动脉狭窄程度相关。
Objective To explore the expression of receptor for activated C kinase 1 ( RACK1 ) in peripheral blood and its relationship with the extent of coronary lesions in patients with coronary artery atherosclerosis. Methods A total of 83 patients with coronary artery atherosclerosis were enrolled. According to the results of angiography, the extent of coronary stenosis was evaluated by the coronary stenosis score. Patients were divided into four groups: group ⅠⅡⅢⅣ (0 score, normal control group), group Ⅱ (1-10 scores); group Ⅲ (11-20 scores), and group Ⅳ ( 〉 20 scores). RACK1 mRNA expression in peripheral blood was detected by real-time polymerase chain reaction. Results The RACK1 mRNA expression in peripheral blood in group Ⅱ , group Ⅲ and group Ⅳ were significantly higher than in group Ⅰ ( t = 10. 62, 12.90, 14.00, respectively, all P 〈 0.05). RACK1 mRNA expression in group Ⅲ and Ⅳ were significantly higher than in group 1] ( t = 9.40, 10. 74, respectively, both P 〈 0. 05 ) , and there was no statistical significance between group Ⅲ and group Ⅳ ( t = 1.43, P 〉 0. 05 ). Correlation analysis showed that RACK1 mRNA expression in peripheral blood was positively correlated with the extent of coronary lesions ( rs = 0. 59, P 〈 0. 01 ). Conclusions The expression of RACK1 is up-regulated in peripheral blood in coronary artery atherosclerosis patients, and it is positively correlated with the extent of coronary lesions.
