荧光定量聚合酶链反应在淋球菌检测中的应用评价 被引量:1

摘要 目的以淋球菌porA假基因为靶序列,建立灵敏度高,且特异性强的荧光定量(FQ)聚合酶链反应(PCR)检测方法。方法以淋球菌porA假基因为靶序列设计引物和探针,在实时荧光PCRMasterMix基础上优化PCR反应体系和方法,并评价方法的灵敏度、特异性和稳定性等。结果经评估表明该法特异性和敏感性均为100.0%,灵敏度较高,达到检测淋球菌小于50CFU/mL的检测限;批间和批内检测变异系数均小于5%。结论以淋球菌porA假基因为靶序列建立FQ-PCR检测方法,灵敏度高,特异性强,且稳定性好,适合于作为淋病临床实验室辅助诊断和淋球菌阳性结果的确证方法。
