摘要
目的研究抑癌基因WWOX在非小细胞肺癌中的蛋白表达及临床意义。方法应用免疫组织化学二步法检测40例非小细胞肺癌石蜡标本及21例癌旁正常肺组织中WWOX蛋白的表达。结果①癌旁正常肺组织中WWOX蛋白阳性表达率高于非小细胞肺癌组织(85.71oAVS37.50oA,P〈0.05),鳞癌中WWOX蛋白阳性表达率与腺癌差异无统计学意义(34.62%VS42.86%,P〉o.05);②男性非小细胞肺癌患者癌组织中WWOX蛋白阳性表达率低于女性非小细胞肺癌患者(25.00%VS56.25%,P〈0.05),吸烟指数≥400的患者癌组织中WWOX蛋白阳性表达率低于吸烟指数〈400的患者(19.05%VS57.890,P〈0.05),而WWOX蛋白呈阳性表达的患者年龄与阴性表达者之间差异无统计学意义(P〉0.05);③分化不良的非小细胞肺癌组织中WWOX蛋白阳性表达率与中等分化和分化良好者相近(P〉o.05);④I+II期非小细胞肺癌患者癌组织中WWOX蛋白阳性表达率与Ⅲ+Ⅳ期患者相比差异无统计学意义(P〉0.05),转移病例的非小细胞肺癌组织中WWOX蛋白阳性表达率与无转移病例相近(P〉0.05)。结论WWOX蛋白在非小细胞肺癌组织中表达较癌旁正常肺组织降低,且与患者性别及吸烟指数密切相关,而与年龄、病理类型、组织分化程度、临床分期以及有无转移无明显关系。
Objective To study the protein expression level of the tumor suppressor gene WWOX in non-small cell lung cancer (NSCLC) and its clinical significance. Methods The protein expression of WWOX gene in paraffin-embedded specimens of 40 cases of NSCLC and 21 cases of adjacent normal lung tissue was detected by two stage method of immunohistochemistry. Results ① The positive rate of WWOX protein expression was higher in adjacent normal lung tissue than that in NSCLC tissue (85.71 vs 37.50%, P 〈0.05) ,and there was no obvious difference in the protein expression of WWOX between squamous carcinoma and adenocarcinoma (34.62% vs 42.86%, P 〉0.05). ②The protein positive rate of WWOX was lower in male patients than that in females (25.00% vs 56.25%, P 〈0.05) ,and the protein expression of WWOX was lower in patients with smoking index≥400 than patients with smoking index〈 400 (19.0% vs 57.89%,, P 〈0.05). There was no significant difference in mean age between the patients whose WWOX protein expression was positive and those whose WWOX protein expression was negative ( P 〈 0.05). ③There was no significant difference in WWOX expression among the poorly, medium and well differentiated cancer tissue ( P 〉0.05). ④The protein expression of WWOX in NSCLC had no significant difference between the patients with stage Ⅰ+ Ⅱ and stage Ⅲ - Ⅳ ( P 〈0.05). Whether there was metastasis or not, the WWOX protein expression in NSCLC was consistent.Conclusions WWOX protein expression in NSCLC tissue is lower than that in adjacent normal lung tissue, and is related to gender and smoking index, but has no obvious relationship with age, pathological type,degree of differentiation, clinical stage and metastasis.
