Ⅱ型糖尿病与非糖尿病大鼠骨质疏松中TGF-β1蛋白表达变化研究 被引量:1

Study on Changes of TGF-β1 Protein Expression in Osteoporosis of Type Ⅱ Diabetes and Non-Diabetic Rats

摘要 目的:观察链尿佐菌素(STZ)诱导糖尿病大鼠与非糖尿病大鼠骨组织中TGF-1蛋白表达及骨密度的变化。方法:将60只Wistar大鼠随机分为对照组、非糖尿病组和链尿佐菌素(STZ)诱导Ⅱ型糖尿病组。取各组大鼠股骨,用免疫组化的方法测定测定股骨中TGF-β1蛋白表达,用XR-26型双能x线骨密度仪测大鼠离体股骨上1/3骨密度。结果:(1)与对照组比较,非糖尿病组和II型糖尿病组血中的GLU、甘油三酯(TG)、低密度载脂蛋白胆固醇(LDL)、总胆固醇(TC)含量明显升高(P<0.05),而Ca2+明显下降(P<0.05);(2)与对照组比较,非糖尿病组和Ⅱ型糖尿病组股骨骨组织中TGF-β1蛋白表达上调(P<0.01)。结论:STZ诱导的Ⅱ型糖尿病模型股骨骨组织中TGF-β1蛋白表达上调,并伴有血脂代谢紊乱,表明Ⅱ型糖尿病可能通过TGF-β1蛋白表达上调来促进股骨骨质疏松病情的发展。 Objective:To observe TGF-β1 protein expression and the changes of bone mineral density in bone tissue of diabetic rats induced by streptozocin(STZ)and non-diabetic rats.Methods:Sixty Wistar rats were randomly divided into control group,non-diabetic group and the streptozocin(STZ)-induced diabetic group.The femur of the rats was taken and TGF-β1 protein expression of the femur was measured with the method of immunohistochemical determination,and then the bone density of the one third of rat femur in vitro was measured with the XR-26 dual energy x-ray absorptiometry.Results:Compared with control group,the GLU,triglycerides(TG),low density lipoprotein(LDL)and total cholesterol(TC)of blood in non-diabetic group and type Ⅱ diabetes had an evident increase(P〈0.05),while Ca2+ had an evident decrease(P〈0.05).Compared with control group,TGF-β1 protein expression of femoral bone tissue had been significantly increased in non-diabetic group and type Ⅱ diabetes group(P〈0.01).Conclusion:There is an evident increase in the TGF-β1 protein expression of femoral bone tissues in STZ-induced type Ⅱ diabetes model,accompanied by dyslipidemia,which indicates that type II diabetes may promote the femur osteoporosis through the increase of TGF-β1 protein expression.
