黄连解毒汤对老年痴呆大鼠SOD活性,MDA含量,Ⅰ-κB和NF-κB蛋白表达的影响 被引量:19

摘要 目的:研究黄连解毒汤(HJD)对老年性痴呆(AD)模型大鼠脑组织的保护作用。方法:Aβ1-42ICV(侧脑室)显微注射建立AD大鼠模型,50只健康SD大鼠随机分为5组,即对照组,Aβ1-42模型组(AD大鼠组),Aβ1-42+HJD低剂量组(5g.kg-1),Aβ1-42+HJD中剂量组(10g.kg-1),Aβ1-42+HJD高剂量组(15g.kg-1)。在Aβ1-42造模7天后,各组均灌胃给药(对照组和AD大鼠组给以生理盐水),1次/d,连续给药21天。采用生物化学方法检测HJD对AD模型大鼠海马组织SOD活性,MDA含量的影响,蛋白印迹检测方法检测I-κB和NF-κB蛋白的表达。结果:与对照组相比,Aβ1-42模型组大鼠海马组织SOD活性降低,MDA含量升高(P<0.01,P<0.05),Ⅰ-κB-NF-κB信号通路活化。HJD作用后,AD模型大鼠海马组织SOD活性升高,MDA含量降低(P<0.05),并抑制Ⅰ-κB-NF-κB信号通路活化。结论:HJD通过提高SOD活性,降低MDA含量,抑制I-κB-NF-κB信号通路活化,改善AD模型大鼠的脑组织生化学改变和炎症改变,对AD有一定的改善和保护作用。 Objective:To study protective effects of Huanglian Jiedu Decoction(HJD) on Alzheimer’s disease model rat’s brain.Methods:The model of Alzheimer’s disease was established with microinjection Aβ1-42 on ICV of rat’s brain.Fifty healthy SD rats were randomly divided into 5 groups: control,Aβ1-42,Aβ1-42+HJD(5g·kg-1),Aβ1-42+HJD(10g·kg-1),Aβ1-42+HJD(15g·kg-1).Five groups rats were treated with HJD or saline solution respectively.After 21 days,the activities of SOD and contents of MDA of rat’s hippocampus tissue were observed by biochemical method.The expression levels of Ⅰ-κB and NF-κB were detected by Western blot.Results:The activities of SOD and contents of MDA in Aβ1-42 group were significantly increased(P〈0.01,P〈0.05) followed by up-regulated expression of NF-κB and down-regulated expression of Ⅰ-κB comparing with control group.After treated by HJD,the activities of SOD were increased,the contents of MDA were reduced(P〈0.05) and the activation of Ⅰ-κB-NF-κB signal pathway was inhibited comparing with Aβ1-42 group.Conclusion: HJD improves the oxidative stress and inflammatory injury of AD model rat’s brain by the mechanism of increasing the activities of SOD,reducing the contents of MDA and inhibiting the activation of Ⅰ-κB-NF-κB signal pathway.
