摘要
从培养液初始pH值、培养温度、摇床转速、培养时间、初始接种量、不同培养基等6个方面对4株石油降解细菌的发酵培养条件进行了初步研究。结果表明:每100 mL培养液中接种0.5 mL种液时,Y-4菌株在30~35℃、pH值7.0~7.5、150 r/min转速条件下培养18 h时产菌量最多,6~24 h为对数生长期;Y-5菌株在35℃、pH值7.3~7.5、150 r/min转速条件下培养24 h时产菌量最多,10~24 h为对数生长期;Y-7菌株在35~37℃、pH值7.0~7.2、150 r/min转速条件下培养30 h时产菌量最多,12~30 h为对数生长期;每100 mL培养液中接种1 mL种子液时,Y-12菌株在35℃、pH值7.9~8.1、150 r/min转速条件下培养36 h时产菌量最多,18~30 h为对数生长期;同时采用4个培养基进行培养,结果表明4个菌株在4种培养基中都能生长,但在营养条件比较丰富的1#培养基中生长较好,在2#、3#、4#培养基中生长较差,4个菌株中Y-7的产菌量最少。
The fermentation conditions of four strains petroleum-degrading bacteria were studied,including the initial pH value of culture solution,raise temperature,shaking speed,raise time,initial inoculum,different medium.The results showed that,when per 100 mL liquid medium inoculated 0.5 mL seed solution,the bacteria amount of Y-4 strain cultured 18 h would be most under the conditions of 30~35℃,pH 7.0~7.5,150 r/min,6~24 h was the logarithmic growth phase;the bacteria amount of Y-5 strain cultured 24 h would be most under the conditions of 35℃,pH7.3~7.5,150 r/min,10~24 h was the logarithmic growth phase;the bacteria amount of Y-7 strain cultured 30 h would be most under the conditions of 35~37℃,pH 7.0~7.2,150 r/min,12~30 h was the logarithmic growth phase;when per 100 mL liquid medium inoculated 1.0 mL seed solution,Y-12 strain cultured 36 h would be most under the conditions of 35℃,Ph 7.9~8.1,150 r/min,18~30 h was the logarithmic growth phase.Four strains were cultured with four different media.The results showed that four strains could grow in the four kinds of medium,and grew well in the 1# nutrient-rich medium,and the growth was poor in the 2#,3#,4# medium,producing bacteria volume of Y-7 strain was the worst.
